Effect of Empagliflozin on Sphingolipid Catabolism in Diabetic and Hypertensive Rats

Abstract: The profile of sphingomyelin and its metabolites shows changes in the plasma, organs, and tissues of patients with cardiovascular, renal, and metabolic diseases. The objective of this study was to investigate the effect of empagliflozin on the levels of sphingomyelin and its metabolites, as well as on the activity of acid and neutral sphingomyelinase (aSMase and nSMase) and neutral ceramidase (nCDase) in the plasma, kidney, heart, and liver of streptozotocin-induced diabetic and Angiotensin II (Ang II)-induced… Show more

“…Male Wistar rats (300–350 g) were divided into two groups (n = 10 each): (1) control and (2) streptozotocin (STZ)-induced diabetic rats. The animals with blood glucose of 300 mg/dl (18 mmol/L) were used as diabetic rats and maintained for 30 days, as we previously reported [ 71 ].…”

“…Male Wistar rats (350–360 g) were divided into two groups (n = 10 each): (1) control and (2) hypertensive rats (435 ng/kg/min of Ang II, which was infused through osmotic minipumps for 14 days (Alzet 2002; Alza, Palo Alto, CA, USA), as we previously reported [ 71 ].…”

“…A single study has shown that ceramide-induced vasodilation is partially endothelium-dependent via activation of NOS3 and subsequent NO generation, as aforementioned, by impairing endothelium function ( 79 ). Angiotensin II (ANGII) may indirectly increase ceramide production, thereby activating SMases in the peripheral vasculature to control vascular function ( 80 ).…”

“…Estas permiten identificar y cuantificar algunos lípidos mediante el uso de anticuerpos monoclonales o de colorantes fluorescentes [100][101][102] . Recientemente reportamos que con la técnica de ELISA (Enzyme-Linked-Immunosorbent Assay) es posible determinar la concentración de ceramida, esfingosina y esfingosina-1-fosfato en plasma y tejidos 103 , lo cual permitirá en un futuro aplicarlo en la clínica.…”
