Ascorbic acid (AA; vitamin C) is an important antioxidant that is directly involved in regulating physiological and psychological systems such as anxiety, memory, depression, stress, and emotional mood states. Environmental enrichment (EE) studies in animals have shown improvements in anxiety and memory as measured by behavioral experiments from living in improved settings. In the present study, the influences of AA and enriched environments were explored in 13-month-old rats, considered middle-aged, half of which were housed for up to 13 months in an enriched environment with a rich diversity of objects to explore, while the other half was maintained in a standard environment with only water and food, to understand better their independent and combined benefits to improving anxiety and memory. The rats were administered either a saline solution or AA (100 mg/kg, orally [p.o.]) 1 hr before being tested on the elevated plus maze (EPM) and the Morris water maze (MWM) behavioral tests. AA reduced anxiety-like behavior and improved spatial memory relative to saline-treated control rats. EE increased the number of entries to the open arms (anxiolytic-like behavior) and improved spatial memory retention. EE potentiated the anxiolytic-like effect of AA. These findings provide additional evidence that AA might play an important psychological therapeutic role as adjunctive psychopharmacotherapy in reducing levels of anxiety and memory deficits in human subjects.

Vitamin C (Vit C) is considered to be a vital antioxidant molecule in the brain. Intracellular Vit C helps maintain integrity and function of several processes in the central nervous system (CNS), including neuronal maturation and differentiation, myelin formation, synthesis of catecholamine, modulation of neurotransmission and antioxidant protection. The importance of Vit C for CNS function has been proven by the fact that targeted deletion of the sodium-vitamin C co-transporter in mice results in widespread cerebral hemorrhage and death on post-natal day one. Since neurological diseases are characterized by increased free radical generation and the highest concentrations of Vit C in the body are found in the brain and neuroendocrine tissues, it is suggested that Vit C may change the course of neurological diseases and display potential therapeutic roles. The aim of this review is to update the current state of knowledge of the role of vitamin C on neurodegenerative diseases including Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, multiple sclerosis and amyotrophic sclerosis, as well as psychiatric disorders including depression, anxiety and schizophrenia. The particular attention is attributed to understanding of the mechanisms underlying possible therapeutic properties of ascorbic acid in the presented disorders.

OBJECTIVE: Methylphenidate (MPH) is a first-line treatment option for attention-deficit hyperactive disorder and narcolepsy. MPH is one of the most abused psychostimulants by the adults and young population to stay awake, perform better, or improve concentration. The scanty reports say that the medical users or abusers mostly consider the administration of benzodiazepines to overcome the adverse effects, i.e., mood- and anxiety-related problems associated with MPH chronic abuse. This work aims to study the effect of alprazolam (ALZ) on MPH-associated adverse effects on liver and kidney. MATERIALS AND METHODS: Female Wistar rats ( n = 58) were administered with MPH (10, 20, and 40 mg/kg) and ALZ (5, 10, and 20 mg/kg) alone and in combination for 28 days. Bodyweight, feed intake, and water intake were monitored weekly. Parameters related to liver and renal function, oxidative stress, and histopathology were performed to evaluate the toxic impacts on the liver and kidneys. RESULTS: ALZ, along with MPH, increased the serum alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, creatinine, and urea levels. The co-abuse also led to elevated oxidative stress and structural abnormalities in the liver and kidney tissues. CONCLUSION: The co-abuse of ALZ has amplified the hepato-renal toxic effects of MPH. Therefore, it is a significant concern for public safety, and their co-abuse must be restricted and discouraged.
