Effect of Donor Age on Morphologic Variation of Cultured Human Corneal Endothelial Cells

Abstract: Cultured HCEC established from older donor tissue display greater heterogeneity. The use of HCEC from younger donors may be preferable to maximize the benefits of HCEC transplantation.

“…Since HCECs can be grown in culture, [35][36][37][38][39][40] the cell-based therapy of using cHCECs to treat corneal endothelial dysfunction has been extensively explored. 11,[41][42][43][44][45] One of the most notable obstacles to the application of cHCECs for cell-based therapy is the lack of an established reliable method for verifying the quality of cHCECs.…”

“…For the preparation of the extracts from bovine corneal endothelium (BCEE), bovine corneal Descemet's membranes were peeled off and the endothelial cells were cultured as described previously [16]. The confluent endothelial cells were collected and resuspended using DMEM/F-12 with 10% FBS and frozen at −20°C for 30 minutes and thawed at 37°C three times.…”
