Effect of Dianhydrodulcitol (DAD) on the Primary Immune Response of Normal and Tumor Bearing Rats

Abstract: The effect of dianhydrodulcithol (DAD) administered in a dose of 5 mg/kg on the number of plaque-forming cells (PFC) in spleens and on the titre of hemolytic antibodies in the sera of rats immunised with sheep red blood cells (SRBC) have been examined. The compound elicited a short-lasting suppressive effect when applied after immunisation; the number of PFC decreased two days after treatment while 5 days after drug administration increased as compared to the untreated animals. Treatment prior to immunisation … Show more

“…The circulating hemolytic antibodies in the individual sera were titrated in Takatsy's microtitrator [2], as de scribed earlier [3], The highest dilutions producing complete hemo lysis and the latest dilutions producing no effect were registered. The titer of 7 S (IgG-typc, ME-resistant) antibodies was determined after digestion of the sera with 0,2 M 2-mercaptoethanol for 30 min at 37 °C [4).…”

“…The circulating hemolytic and hemagglulinating antibodies of individual sera were titrated in Tak&tsy's microtitrator [16] as de scribed earlier [5], The lowest dilutions producing no lysis or ag glutination were recorded. The t tcr of 7-S (IgG-type.…”

“…The drug proved to be effective and well tolerated also in humans. DAG had a suppressive effect on the immune system inhibiting the in vitro cytostatic activity of lym phocytes [14] and the humoral immune responsiveness of rats [5], Alterations in the immunomodulating ef fect depending on dose or schedule of treatment were described with numerous cytostatic agents [3,8,10]. This work was done to clarify to what extent the dose and time of DAG treatment would influence the hu moral immune response to sheep red blood cell (SRBC) antigen.…”
