Effect of cycloheximide and tunicamycin on the gonadotrophin-releasing hormone stimulated distal glycosylation of luteinizing hormone by rat pituitary cells

Abstract: We have previously demonstrated that gonadotrophin-releasing hormone (GnRH) induces not only changes in quantity but also in quality on secreted luteinizing hormone (LH), by increasing [14C]Leu (translation) and [3H]Gal (distal glycosylation) incorporation into newly synthesized hormone. In the present report, we have further examined the GnRH-induced [3H]Gal-LH synthesis and release by treating anterior pituitary cells with polypeptide synthesis and glycosylation inhibitors (cycloheximide and tunicamycin, res… Show more

“…In contrast, the present results show the relative increase in less alkaline LH species in the proestrus-afternoon pituitaries. These findings are consistent with those of our previous studies [11,12] showing that GnRH increased [ 3 H]galactose incorporation, by acting directly at the distal glycosylation level. In the case of pituitaries from proestrusafternoon rats, the shift toward isoforms with pI values in the range of 9.00-8.01 could potentially result from sialylation of increased terminal galactose.…”

“…In our previous studies [11,12] we presented evidence of the GnRH-stimulatory effect on terminal LH glycosylation in vitro as estimated by increased [ 3 H]Gal incorporation into newly synthesized hormone. On the basis of those findings, the aim of the present report was to determine possible modifications in pituitary LH isoforms induced by the in vivo increase in GnRH at the time of the preovulatory gonadotropin surge.…”

“…Tunicamycin (TM), an inhibitor of protein glycosylation, was used to inhibit protein folding and consequently forced the degradation of the protein by any available ERAD pathway. Tunicamycin has been shown to effectively inhibit protein glycosylation without affecting protein synthesis [ 28 , 29 ]. Inhibitors of ERAD I or ERAD II pathways were used in combination with TM to block protein degradation, and then the variation of the intracellular rht-PA quantity was observed.…”
