Effect of a monoclonal antibody against interleukin‐4 on collagen‐induced arthritis in mice

Abstract: 1 The present study was undertaken to investigate the eect of a monoclonal antibody (11B11 mAb) against interleukin-4 (IL-4) on collagen-induced arthritis (CIA) in mice. 2 11B11 mAb was daily injected intraperitoneally over a period of 10 days, commencing on the day of immunization with type II collagen (CII).3 The results showed that the anti-IL-4 mAb markedly augmented both the incidence and the severity of CIA. The augmentation of the disease was associated with a signi®cant increase in anti-CII IgG2a antib… Show more

“…Similarly, in CIA, injections of anti-IL-4 i.p. for 10 days initiated on the day of immunization with type II articular cartilage collagen markedly augmented the incidence and severity of arthritis (25). However, anti-IL-4 Ab did not affect CIA incidence or macroscopic arthritis evaluation in a similar study (26).…”

“…Prophylactic treatment with IL-4 mAbs was able to significantly reduce the incidence and clinical score [151]. Interestingly, IL-4 À/À mice were only protected from CIA when immunised with collagen in incomplete Freund's adjuvant.…”

“…Th1 cells, which produce interferon-(IFN-), have been considered to be the major player in CIA. In fact, spleen cells from CIA mice produce a large amount of IFN-after antigen stimulation in vitro [3]. Recently, interleukin-17 (IL-17)-producing Th17 cells have been identiWed, and recent studies have suggested that Th17 cells play a central role in the pathogenesis of autoimmune disease, because neutralization or genetic deletion of IL-17 inhibits the development of disease in several autoimmune disease models including CIA model [4,5].…”
