Histomorphometric and Histopathologic Evaluation of the Effects of Systemic Fluoride Intake on Orthodontic Tooth Movement

Objectives The aim of this study was to determine the effects of systemic fluoride intake on orthodontic tooth movement with histomorphometric and histopathologic methods. Materials and Methods Forty-eight Wistar albino rats were randomly divided into four groups of 12 rats each. Group I received fluoridated water and underwent orthodontic tooth movement. Group II received fluoridated water and did not undergo orthodontic tooth movement. Group III received nonfluoridated water and underwent orthodontic tooth movement. Group IV received nonfluoridated water and did not undergo orthodontic tooth movement. At the beginning of the experiment (T1), impressions were taken from the maxilla of the rats in groups I and III under general anesthesia, and a NiTi closed coil spring appliance was ligated between the left maxillary central incisors and maxillary first molar. The orthodontic force applied was approximately 75 g, and the duration of the experimental period was 18 days. During the experimental period, appliances were controlled daily. At the end of the experimental period (T2), the rats were sacrificed with an overdose of a ketamine/xylasine combination, and their impressions were obtained. The upper first molars were subsequently dissected for histological examination. Incisor–molar distance, number of osteoblasts, number of osteoclasts and periodontal ligament (PDL) space widths on the compression and tension sides were measured. Statistical Analysis All measurements were statistically analyzed with SPSS for Windows version 18.0 (SPSS Inc., Chicago, IL, USA). Repeated measures ANOVA and posthoc Tukey tests were used to compare the groups. Results No statistically significant difference was found with respect to the amount of tooth movement between the fluoridated and nonfluoridated groups (p > 0.05). Orthodontic force application increased the number of osteoblasts at the tension sides and reduced it at the compression sides (p < 0.001). An increased number of osteoclasts was observed in the nonfluoridated group relative to the fluoridated group (p < 0.01). Conclusions No difference was observed with respect to the amount of tooth movement between the fluoridated and nonfluoridated groups. Fluoride significantly reduced the number of osteoclasts in the experimental groups.

This study was conducted to determine the influence of long-term tablet fluoridation on primary pulp calcification by light microscopy. Twenty-four caries-free primary molars (after continuous postpartally initiated 1- to 10-year tablet fluoridation) were compared to 17 primary molars of children without fluoride prophylaxis. Pulp calcification in children with tablet fluoridation was significantly more frequent and more pronounced than in untreated children (p = 0.001). Besides the known pulp stones, the prophylaxis group evidenced a special form of calcification consisting of fibrodentin-like hard tissue not observed in the untreated children. These hard tissue bodies developed "intramurally" on the pulp floor and the inside of the dental roots with an irregular extramural spread into the coronal and radicular pulp by displacement and fibrotization of the pulp tissue. Moreover, some of the teeth had more or less extensive areas of interglobular dentin. The affected teeth were ankylosed in the area of the bi- and trifurcation and on the inside of the roots and were thus infra-occluded. Although the duration of tablet fluoridation has no statistically significant influence on pulp calcification, there is a correlation between extensive pulp calcification, postnatally initiated fluoride prophylaxis and the infraocclusion of primary molars.

Objective: This study aims to investigate the influence of fluoride exposure on implant osseointegration.Methods: A total of 24 male New Zealand white rabbits were randomly divided into the control group and the fluoride exposure group. Rabbits in the control group were fed with tap water, while those in the fluoride exposure group were given 200 mg/L sodium fluoride solution. After 2-month feeding, implants were inserted into the extraction socket immediately after extraction of rabbit mandibular anterior teeth.Four rabbits in each group were sacrificed to collect the implants samples at 1, 2, and 3 months post-implantation, respectively. Radiographic and histomorphometry examinations were performed to evaluate the condition of implant osseointegration.Results: Bone volume around the implants increased in a time-dependent manner in both groups. Micro-CT images illustrated that the bone mineral density (BMD) in the fluoride exposure group was significantly lower than that in the control group after implantation for 2 and 3 months. The bone-implant contact ratio (BIC) in the fluoride exposure group was much lower than that of the control group at 3 months post-implantation according to histomorphometry examination. Conclusions:In rabbit animal model, high fluoride exposure affected the quality of bone surrounding the implant and significantly reduced bone integration of the implant, especially in the late stage of osseointegration.
