Eduscho Coffee Extract Effectively Inhibits the Formation of Amyloid-like Fibrils by Trypsin in Aqueous Ethanol

Abstract: In this work we used an in vitro trypsin aggregation model to show that certain commercial coffee extracts can inhibit protein aggregation. Aggregation experiments were performed using several spectroscopic methods and a dye binding assay, such as turbidity, Congo red (CR) and electronic circular dichroism (ECD), that was further supported by transmission electron microscopy (TEM). A correlation was found between the anti-aggregation properties and the total phenolic content of the coffee extracts. The results… Show more

“…Ethanol (EtOH) is the most commonly used alcohol in beverages available in the market with the highest percentage varying from 3–50 % (v/v) [13] . Aggregation of Trypsin is triggered in the presence of 60 % EtOH as reported by Kotormán and co‐workers [14–17] . Different mutations and inactivation of genes due to autodigestion of pancreatic Trypsin, triggered by alcohol consumption, serve as the active precursor of pancreatitis and pancreatic cancer [18] .…”

“…19 A number of studies have shown that polyphenolic compounds inhibit the fibril formation. [20][21][22][23][24] The antihypertensive, lipid-lowering, antiobesity, antidiabetic, antiatherosclerotic, antithrombotic, and cardioprotective effects of avocado have been demonstrated in many studies. 16 The avocado fruit phytochemicals also can provide a beneficial nutrition strategy for cancer prevention.…”

“…Phenylmethylsulfonyl-trypsin amyloid-like fibrils were prepared as previously reported by Kasi et al 31 Earlier we had showed that PMS-trypsin forms amyloid-like fibrils in 60% ethanol/10 mM phosphate buffer (pH 7.0). 32 The detection of amyloid growth is usually done by measuring the turbidity of the solution.…”

“…16 Polyphenols are structurally characterized by 2 or more hydroxyl groups attached to 1 or more benzene rings. 17 Many polyphenolic compounds have been shown to inhibit significantly the formation of amyloid fibrils in vitro , 18,19 because the aromatic polyphenolic rings of polyphenols and proteins may associate via π-π stacking interactions with each other. 20 They also block the self-assembly process of the protein.…”
