Early Manifestations in Multiple-low-dose Streptozotocin-induced Diabetes in Mice

Abstract: This study suggests that transferred or cocultured MSs from mld-SZ mice exert a functional immune aggression against beta cells at a very early stage, before donor mice develop impaired insulin secretion and hyperglycemia.

“…These data are in agreement with previous immunological and morphological findings. 7 Our results show a clear interrelationship between lipids and insulin secretion in the early stage of its evolution. Impact of changes in lipids on the β-cell metabolism and viceversa has been clearly suggested in this paper.…”

“…However, hyperglycemia and diminished insulin secretion from perifused pancreatic islets only appeared at days 8 and 9 after mld-STZ. 7 Moreover, Ebeling et al 8 recently showed that type 1 diabetes is associated with an increase of intramuscular triglyceride content. Patients with type 1 diabetes have elevated basal lipid oxidation rates, an increased availability of plasma free fatty acids (FFa) seems to play a role in the early stages of experimental type 1 diabetes mellitus induced in c57BL/6J mice by multiple low doses of streptozotoxin (mld-sTZ).…”

“…As a consequence, immune cells respond to ␤-cell self-antigens and the progression of hyperglycemia and insulitis evolve during a 2-week period (Cvjetićanin et al, 2010). This MLDS model has been largely employed to study the immunological mechanisms underlying diabetes, including its immunopathogenesis and also therapeutic strategies (Karabatas et al, 2005;Kanitkar et al, 2008).…”

“…Diabetes was induced by multiple low dose (40 mg/kg/day) administration of streptozotocin (STZ), for 5 consecutive days [29]. Six groups were considered as silymarin-treated (Sil) groups, in which silymarin was administered in three doses of 20 (Sil-20), and 80 (Sil-80) mg/kg.…”
