E6/E7 and E6* From HPV16 and HPV18 Upregulate IL-6 Expression Independently of p53 in Keratinocytes

Abstract: Keratinocyte infection with high-risk human papillomavirus genotypes has been linked to cancer development. In cervix, the alpha HPV16 and HPV18 have been reported as the mayor causative agents of cervical cancer. Oncogenic progression and chronic inflammation are closely related processes, with IL-6 as one of the main pro-inflammatory cytokines involved. However, there are limited studies about the regulation of IL-6 by low and high risk HPVs and the HPV proteins implicated in this modulation. In this work, w… Show more

“…Several studies have used this cell line as a model to address the effect of the HPV E6 and E7 oncogenes in different processes associated with cell transformation and immune evasion. (27,42,43) Interestingly, HaCaT cells harbour a p53 gene mutational spectrum that is typical of ultraviolet light-induced mutations. (44) This p53 form is not degraded by the ectopic expression of HPV E6.…”

“…Indeed, as we already described before, the E6 and E7 HPV oncoproteins are involved in cell transformation and carcinogenesis and have been proven to be indispensable for maintenance of tumor phenotype (32). Moreover, recent in vitro data suggest that E6 and E7 oncoproteins and spliced isoforms of E6 oncoprotein would be associated with higher levels of IL6, responsible of an immunosuppressive environment within cancer (114). This immunosuppressive context could be targeted by therapies associating IL6 and PD-1/PD-L1 blockade (115).…”

“…We next evaluated the selectivity index (SI) for these species based on the response (IC 50 value) of a normal human skin keratinocyte cell line, HaCaT. HaCat cell line is commonly used to represent in vitro human papillomavirus (HPV) infection-induced carcinogenesis [ 18 ]. The analysis showed selectivity (SI higher than two) for A. crassiflora and S. adstringens in CaSki and HeLa cell lines ( Table 2 ).…”
