Dynamics of receptor-operated Ca2+ currents through TRPC channels controlled via the PI(4,5)P2-PLC signaling pathway

Abstract: Transient receptor potential canonical (TRPC) channels are Ca2+-permeable, nonselective cation channels that carry receptor-operated Ca2+ currents (ROCs) triggered by receptor-induced, phospholipase C (PLC)-catalyzed hydrolysis of phosphatidylinositol 4,5-bisphosphate [PI(4,5)P2]. Within the vasculature, TRPC channel ROCs contribute to smooth muscle cell depolarization, vasoconstriction, and vascular remodeling. However, TRPC channel ROCs exhibit a variable response to receptor-stimulation, and the regulatory … Show more

“…Calcium ion uptake by cells is related to voltagedependent calcium ion channels (23), receptor-operated calcium ion channels (24) or calcium ion release from the endoplasmic reticulum (25) following activation of PI3K (26), serine-threonine kinase (27), phospholipase A2 and phospholipase C (24,28) or calcium ion/ calmodulin-dependent protein kinase IIA (29). We found that the vacuolation induced by rCAMP factor in RAW cells is inhibited by the PI3K inhibitors LY294002 and wortmannin.…”

“…Among the TRP channels, the TRPC family was indeed a perfect candidate to form ROCE; TRPC2, 3, 6, and 7 can be activated by DAG, a potent lipid messenger produced from PIP 2 by PLC activation, and activation of TRPC4 and 5 was PLC-dependent [52]. In mouse airway epithelial cells, TRPV4 participated in ROCE instead of SOCE [27], further indicating the possible involvement of TRP channels in ROCE.…”

“…We observed that blackcurrant juice inhibited the BaCl 2 -induced contraction of the ileum. Barium causes the depolarization of smooth muscle, which results in the opening of voltage-dependent calcium channels and the entry of calcium ions into cells [30].…”

“…High concentrations of potassium ions are known to cause tonic contraction of the smooth intestinal muscles, causing the opening of voltage-dependent L-type calcium channels, allowing the entry of calcium ions into the cell [30]. Therefore, substances that inhibit contractions induced by high K + concentrations are considered calcium channel blockers [31].…”
