Dynamic and Differential Expression of Duplicated Cxcr4/Cxcl12 Genes Facilitates Antiviral Response in Hexaploid Gibel Carp

Abstract: Chemokine receptor cxcr4 and its ligand cxcl12 have evolved two paralogs in the teleost lineage. In this study, we have identified four duplicated cxcr4 and cxcl12 genes from hexaploid gibel carp, Carassius gibelio, respectively. Cgcxcr4bs and Cgcxcl12as were dynamically and differentially expressed in immune-related tissues, and significantly up-regulated in head kidney and spleen after crucian carp herpesvirus (CaHV) infection. Blocking Cxcr4/Cxcl12 axis by injecting AMD3100 brought more severe bleeding symp… Show more

“…Luciferase activity assays were performed as described previously ( 49 , 50 ). CAB cells were seeded in 24-well plates overnight and co-transfected with various plasmid constructs at a ratio of 10:10:1 (promoter-driven luciferase plasmid/expression plasmid/Renilla luciferase plasmid pRL-TK) using FuGENE HD Transfection Reagent (Promega).…”

“…Mammalian CXCR4 has been suggested as a marker for an initial common lymphoid progenitor in mice [56] and for differentiated plasmablasts or plasma cells both in mice and humans [57,58]. Several studies have analyzed the evolution and function of cxcr4 genes in teleost fish showing the presence of either two paralog genes designated as cxcr4a and cxcr4b [59][60][61] or even four gene copies in cyprinids [62]. Our results identified four different genes annotated as rainbow trout CXCR4-like, which all seemed expressed in B cells.…”
