Dual DNA binding specificity of a petal epidermis-specific MYB transcription factor (MYB.Ph3) from Petunia hybrida.

Abstract: The MYB.Ph3 protein recognized two DNA sequences that resemble the two known types of MYB DNA binding site: consensus I (MBSI), aaaAaaC(G/C)‐GTTA, and consensus II (MBSII), aaaAGTTAGTTA. Optimal MBSI was recognized by animal c‐MYB and not by Am305 from Antirrhinum, whereas MBSII showed the reverse behaviour. Different constraints on MYB.Ph3 binding to the two classes of sequences were demonstrated. DNA binding studies with mutated MBSI and MBSII and hydroxyl radical footprinting analysis, pointed to the N‐term… Show more

“…Second, CCAl probes can only detect a single gene on a DNA gel blot even at low stringency and using a probe including the myb-related sequence. In addition, the DNA region to which it binds, and which is conserved in a number of plant Lhcb genes, does not include the plant consensus sequence CCT/AACC for recognition by the maize Myb P protein (Grotewold et al, 1994), the recognition sequence for other plant Myb proteins (e.g., see Solano et al, 1995), or consensus sequences determined for Myb proteins from other kingdoms (TAACTG) (Tanikawa et al, 1993). We conclude, therefore, that CCAl is a nove1 myb-related gene rather than a member of the myb gene family in Arabidopsis.…”

“…Gene encoding Chalcone synthase (CHS) was activated by another MYB protein from Petunia, PhMYB3, which was expressed specifically in petal epidermis where anthocyanin pigment was made (Solano et al 1995). MYB protein also served to regulate other branches of phenylpropanoid metabolism.…”

“…The oligonucleotide mixtures (RP or ACP) were made double-stranded by annealing with primer F (5Ј-CGC-GAA-TTC-TGC-AGG-ATC-C-3Ј) and using the Klenow fragment of DNA polymerase I to synthesize the other strand. Primers F and R (5Ј-TCG-ACT-CGA-GTC-GAG-ATC-G-3Ј) were used for PCR labelling and amplification during random binding site selection (RBSS) as described in Solano et al (1995).…”
