Drugs modulating the L-arginine:NO:cGMP pathway – current use in therapy

Abstract: Nitric oxide (NO) is a relatively novel messenger that plays a significant role in a wide range of physiological processes. Currently, it is known that, both, lack and excess of NO can cause diseases, thus a lot of substances have been discovered and utilized which can change the concentration of this molecule within the organism. The aim of the present work is to provide an overview of currently used agents modulating the L-arginine:NO:cGMP pathway, as well as to summarize current understanding of their pharm… Show more

“…First nebivolol improves endothelial dysfunction that causes endothelial cells to initiate inflammation by the generation of reactive oxygen species and proinflammatory cytokines [ 22 ]. Second nebivolol increases the level of nitric oxide in vessel causing vasodilatation by interacting with L-arginine/nitric oxide pathway as inflammation blocks the ability of endothelium to generate nitric oxide and prostacyclin [ 23 ]. Finally nebivolol itself has strong antioxidant and anti-inflammatory potential by reducing the production of hs-CRP, interleukin-6 (IL-6), vascular adhesion molecule (VCAM-1), adiponectin, and leptin in hypertensive patients in various clinical studies [ 12 24 ].…”

“…Nitrate medications (nitroglycerin, sodium nitroprusside, isosorbide derivatives, etc.) are pharmaceutical agents with vasodilator effect (Polakowska et al, 2016 ). Thus, medication with isosorbide nitrates as active compounds is often used to prevent angina attacks and in the prophylactic treatments of angina pectoris (Yao et al, 2015 ).…”

“…NG is an organic nitrate that reacts with thiols and other reducing substrates [11]. It is converted into 1, 2-glyceryl dinitrate and nitrite in the vascular smooth muscle cells, which is then metabolized by mitochondrial aldehyde dehydrogenase 2 with a half-life of 1-4 min [13,14].…”
