The absorption and elimination of oxprenolol after intravenous and oral administration has been studied in six healthy male volunteers.The plasma concentration/time curve after intravenous administration is described by an equation consisting of three exponential terms. The terminal half-life of 4.5 hours is longer than previously reported and is consistent with the observed effect on heart rate after a single dose.The bioavailability of oxprenolol has been estimated from a comparison of the area under the curve values after oral and intravenous administration, and shown to be 25 % and 30 % for 80 mg oxprenolol in tablet and solution form respectively. Treatment of the volunteers with either metoclopramide which decreases gastric retention and increases intestinal motility, or neomycin which produces a deterioration in the integrity of the intestinal mucosa, did not appear to alter the absorption of oxprenolol.
