Klebsiella oxytoca, a member of the Enterobacteriaceae, is a gram-negative pathogenic bacterium of environmental origin, which can cause infection in healthcare settings. Outbreaks of multidrug-resistant K. oxytoca infection have been increasingly reported in hospitalized patients. Despite the growing importance of this pathogen, there is limited knowledge about the population structure and epidemiology of antimicrobial resistant K. oxytoca. We investigated the population structure and genomic basis of antimicrobial resistance of 41 multidrug resistant K. oxytoca isolates recovered from bloodstream infections across the UK and Ireland. Our results show that K. oxytoca has a highly diverse population, which is composed of several distinct clades, and we identified one recent expansion of a clone within our dataset. Although the K. oxytoca genomes are clearly distinct from the genomes of a global collection of Klebsiella pneumoniae complex, pre-dominantly composed of K. pneumoniae, we found evidence for sharing of core genes through recombination, as well as the exchange of accessory antimicrobial resistance and virulence factor genes between the species. Our findings also suggest that the different K. oxytoca clades have acquired antimicrobial resistance and virulence factor genes independently. This highlights the clinical and therapeutic importance of genetic flexibility in K. oxytoca and the relevance of this in its role as an opportunistic pathogen.

Microbiological monitoring of consumer products and the efficiency of early warning systems and outbreak investigations depend on the rapid identification and strain characterisation of pathogens posing risks to the health and safety of consumers. This study evaluates the potential of three rapid analytical techniques for identification and subtyping of bacterial isolates obtained from a liquid hand soap product, which has been recalled and reported through the EU RAPEX system due to its severe bacterial contamination. Ten isolates recovered from two bottles of the product were identified as Klebsiella oxytoca and subtyped using matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI TOF MS), near-infrared Fourier transform (NIR FT) Raman spectroscopy and Fourier transform infrared (FTIR) spectroscopy. Comparison of the classification results obtained by these phenotype-based techniques with outcomes of the DNA-based methods pulsed-field gel electrophoresis (PFGE), multi-locus sequence typing (MLST) and single nucleotide polymorphism (SNP) analysis of whole-genome sequencing (WGS) data revealed a high level of concordance. In conclusion, a set of analytical techniques might be useful for rapid, reliable and cost-effective microbial typing to ensure safe consumer products and allow source tracking.

The aim of the study was to assess the effect of N-dodecyl-N-(propylpiperidinium-3-sulfonate) on the physico-chemical and functional properties of liquid soaps for sensitive skin. The studied materials consisted of originally formulated liquid soaps containing amphoteric surfactants including cocamidopropyl betaine and newly synthesized sulfobetaine in the following ratios: 0 : 0, 1 : 1, 0 : 2 and 2 : 0. The aim of study was to examine the effects of the additive on the surface tension of aqueous soap solutions, wetting properties, dynamic viscosity, emulsification of fatty soils, foaming properties, antimicrobial characteristics and skin irritation potential. It is shown that the liquid soaps obtained in the study effectively reduce the surface tension of water – which may indicate good washing properties – and demonstrated desirable wetting properties. The application of N-dodecyl-N-(propylpiperidinium-3-sulfonate) in cosmetic formulations makes it possible to obtain products with the required viscosity, fat emulsification capacity and foaming properties. It needs to be emphasized that sulfobetaines added to the prototypical liquid soaps also demonstrate antibacterial characteristics and reduce the skin irritant effect caused by anionic surfactants.
