Doxorubicin Induced Nephrotoxicity: Protective Effect of Nicotinamide

Abstract: Introduction. Nephrotoxicity is one of the important side effects of anthracycline antibiotics. The aim of this study was to investigate the effects of nicotinamide (NAD), an antioxidant agent, against nephrotoxicity induced by doxorubicin (DXR). Methods. The rats were divided into control, NAD alone, doxorubicin (20 mg/kg, i.p.) and DXR plus NAD (200 mg/kg, i.p.) groups. At the end of the 10th day, kidney tissues were removed for light microscopy and analysis. The level of tissues' catalase (CAT), glutathion… Show more

“…Doxorubicin and Cisplatin are widely used anticancer drugs for various cancers including brain cancer. A large number of reports have documented the hepatotoxicity and nephrotoxicity of these drugs (36)(37)(38)(39). Therefore, alternative therapeutic agents that kill cancer cells without or with low hepatotoxicity and nephrotoxicity are highly desirable.…”

“…Adriamycin toxicity limits its application in chemical therapy ADR has toxic action on the kidneys, thus causes acute renal failure that is a limiting factor for its beneficial application (Ayla et al, 2011). In this work, ADR significantly increased blood urea and creatinine after treatment for 8 weeks.…”

“…The high levels of NO react with superoxide anions, resulting in the formation of peroxynitrite. Peroxynitrite is a potent and aggressive cellular oxidant [36,37]. The increased NO levels in the DOX group (Group 3) could be explained by this mechanism.…”
