Doxorubicin-Conjugated Platinum Theranostic Nanoparticles Induce Apoptosis via Inhibition of a Cell Survival (PI3K/AKT) Signaling Pathway in Human Breast Cancer Cells

Abstract: Herein, we report facile theranostic platinum nanoparticles (PtNPs) conjugated to an anticancer drug, doxorubicin (DOX), in unraveling the inhibition of a cell survival PI3K/AKT (phosphatidylinositol 3-kinase/protein kinase B) signaling pathway in MCF-7 and MDA-MB-231 human breast cancer cells. The significant features of our DOX@PtNPs as a theranostic platform are as follows: (i) drug release studies showed a progressive pH-dependent delivery; (ii) in vitro studies of DOX@PtNPs displayed a relatively higher c… Show more

“…MDA-MB-231 triple-negative breast cancer cells were purchased from the National Centre for Cell Science, Pune, India. The cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM) supplemented with foetal bovine serum ( φ (FBS)=10%) and 1% ( m / V ) antibiotics (penicillin/streptomycin) at 37 °C in a humidified atmosphere with 5% CO 2 and 95% oxygen ( 17 ).…”

“…Metallic and metal oxide NPs can be produced and modified with a variety of chemical functional groups as needed. They can be conjugated with biological molecules (antibodies, nucleic acids or peptides), targeting ligands, and anticancer drugs using appropriate functionalization strategies ( Fu et al, 2020 ; Oliveira et al, 2020 ; Patel et al, 2020 ; Zeng et al, 2020 ). Herein, we summarize the preparation, characterization, and anti-cancer effects of platinum nanoparticles (PtNPs).…”

“…Noble metal based nanoparticles, such as palladium, platinum, gold, and silver have been studied in the field of medicine and pharmaceuticals due to their therapeutic potential. [1][2][3][4][5][6][7] In particular, platinum nanoparticles (PtNPs) have been reported with promising anticancer results. [8][9][10][11][12][13][14] There is evidence of PtNPs' capacity to enter the cell and damage DNA by releasing platinum ions.…”
