Downregulation of OCT4 promotes differentiation and inhibits growth of BE (2)-C human neuroblastoma I-type cells

Abstract: Abstract. In in vitro continuous culture, N (neuroblastic)-, S (substrate adherent)-and I (intermediate)-type cells were identified in human neuroblastoma (NB), where I-type is recognized as a stem cell type. Octamer-binding protein 4 (OCT4) is a cell marker used to identify the stemness of cells, whose roles in regulating I-type NB cells have yet to be fully elucidated. In the present study, we assessed the differentiation regulation role of OCT4 in BE (2)-C of I-type cells. We demonstrated that downregulatio… Show more

“…Multiple studies have documented the aberrant expression of OCT TFs in various types of cancer [5058] including GBM [30, 5963], and the enrichment of this group of TFs in a subpopulation of undifferentiated cancer stem cells indicated their critical role in tumor initiation, metastasis and resistance to chemotherapy. Expression of OCT4 was significantly higher in several malignancies and positively correlated with poor prognosis [6466].…”

“…Previous studies have described Oct4 expression in different human neuroblastic cell lines and side-population cells in NBTs, linking its presence with the stem cell-like phenotype in these tumours [2325]. The characteristic phenotype implies a stem cell self-renewal capacity, but also the presence of tumour cells with multipotential differentiation properties that can be induced to differentiate in neuroblastic or glial phenotypes.…”

“…The overexpression of POU5F1 in BE(2)-C human NB type I cells can promote cell proliferation and increase its colony formation rate [ 29 ]. There are many mechanisms for Pou5f1 to regulate the cell cycle.…”
