Controlling melanogenesis is important for maintaining the good health and cosmetic appearance of a human body. This study aims to search the active compounds from natural products exhibiting the melanogenesis modulating activity and elucidate the mechanism underlying the observed activity. Two novel quercetin glycosideswere isolated and identified from Helminthostachys zeylanica roots 50 % ethanol extract. Compound 1 exhibited intracellular melanogenesis stimulatory activity, while 2 showed no effect even the structural similarity. To understand the structure-activity relationships, twelve quercetin glycosides and seven methylquercetins were synthesized from rutin as starting material. As the result of bioassay using synthesized nineteen quercetin derivatives in B16 melanoma cells, some quercetin-3-O-b-D-glucopyranosides stimulated the intracellular melanogenesis. On the other hand, synthesized 3-O-methylquercetin 12 and 3,4 0 ,7-Otrimethylquercetin 15 increased both intra and extracellular melanin contents with no cytotoxicity. Compoud 15 increased the phosphorylated p38 mitogen-activated protein kinase (MAPK) and microphthalmia-associated transcription factor (MITF) which regulates the expression of tyrosinase, TRP-1 and TRP-2. While 12 enhanced the expression of the melanogenic enzymes without involving the MITF, as evidenced by its lack of any stimulation of the expression of MITF and p-p38 MAPK. This result indicates that 12 may stimulate the expression of tyrosinase, TRP-1, and TRP-2 by stimulating currently unidentified transcriptional factors and/or by regulating the degradation of melanogenic enzymes.

Abstract:With the public's growing interest in skin whitening, lightening ingredients only used under dermatological supervision until recently, are more and more frequently incorporated into cosmetic formulas. The active agents that lighten skin tone are either natural or synthetic substances, and may act at various levels of melanogenesis. They are used to treat various skin pigmentation disorders or simply to obtain a lighter skin tone as whiter skin may be synonymous of wealth, health, youth, and/or beauty in different cultures. However, recent studies demonstrated the adverse effects of some of these ingredients, leading to their interdiction or restricted use under the European Directive and several other international regulations. After an overview of skin whitening practices and the associated risks, this article provides insight into the mechanisms involved in melanin synthesis and the biological assays available to attest the lightening activity of individual ingredients. The legislation dealing with the use of skin lighteners is then discussed. As traditional depigmenting agents such as hydroquinone and corticosteroids are of safety concern, the potential of natural extracts has been investigated more and more; finally, a synthesis of three years of research in our laboratory for such plant extracts will be given.

Hyper pigmentary problems, including post inflammatory hyper pigmentation, solar lentigos, and melasma, occur widely in the human population and are thus of broad interest for control. Tyrosinase is the key enzyme of melanogenesis and it's over expression results in hyper pigmentation. Tyrosinase is not only liable to human pigmentation, but also responsible for the undesirable enzymatic browning of fruits and vegetables, which is of economic importance. So in order to get rid of the problem, inhibitors of the enzyme tyrosinase have been concerned. Advances in genomics and proteomics have opened up new possibilities for the rapid functional assignment and global characterization of proteins and DNA. Organized collection of molecules on arrays have provided a vigorous platform for rapid screening, lead discovery and eradication of molecular characterization. Nowadays, it is used for the discovery of tyrosinase inhibitor. It involves microarrays of immobilized distinct small molecules or DNA or RNA of cells treated with molecules to be tested and binding of tyrosinase proteins or TYR gene respectively. Compound is then screened and the respective compound is identified as a new small-molecule binder to tyrosinase and used it as potential tyrosinase inhibitor. Its actual binding and inhibitory effects on tyrosinase is also validated using an enzyme-based inhibition assay or other binding assays. As a rapidly maturing technology, high-throughput proteomic and genomic pave the way forward in the treatment of skin hyper pigmentation. Hence in the present review we have focused on the use of microarray technique over the conventional strategies for the elucidation of tyrosinase inhibitors.
