Double‐stranded DNA stereoselectively promotes aggregation of amyloid‐like fibrils and generates peptide/DNA matrices

Abstract: An amyloidogenic LAM-L peptide (AASIKVAVSADR, all-L configuration) derived from laminin promoted cell adhesion, neurite outgrowth, and angiogenesis. Here, we prepared novel matrices using double-stranded DNA and the LAM-L peptide. Double-stranded DNA promoted aggregation of amyloid-like fibrils and generated a LAM-L/DNA matrix through electrostatic interactions between the phosphate groups of DNA and the amino groups of LAM-L. This formation of peptide/DNA matrix depends on the Ile-Lys-Val-Ala-Val (IKVAV) sequ… Show more

“…[29][30][31][32], was shifted to a lower wavenumber with the accumulation of L-Asp and D-Asp (see the dashed line in Figure 2). This is due to the electrostatic interaction between the amino group of Asp and the phosphate group of DNA, and a similar interaction has been reported for the DNA-peptide biomatrix [21] and DNA-poly(allylamine) composite material [33]. Additionally, the absorption band at 1533 cm −1 , related to the in-plane vibration of the cytosine (C) base and the stretching vibration of C = N of the guanine (G) base [29][30][31], was shifted to a higher wavenumber when the DNA-film accumulated the L-Asp (see the dashed line in Figure 2).…”

“…We also recently reported the stereo-selective aggregation of an amyloidogenic peptide (AASIKVAVSADR) by the addition of double-stranded DNA [21]. Consequently, double-stranded DNA promoted aggregation of the amyloidogenic peptide, which consisted of only the L-form amino acid, and the amyloidogenic peptide/DNA matrix indicated a strong cell attachment activity [21].…”

“…In fact, the similar phenomena, such as the shift to a lower wavenumber of phosphate group by the electrostatic interaction, have been reported at various hybrid materials with the electrostatic interaction. 12,14,20 In contrast, the dried SiNSi material showed the absorption band at 1000-1200 cm À1 , 20,37,38 have been reported for various organic-inorganic hybrid materials with the silane coupling reagents. 20,38 Generally, the methoxysilyl group in SiNSi molecule hydrolyzes in water and produces the silanol group.…”

“…[1][2][3][4] Therefore, double-stranded DNA has been used as a functional material, such as the accumulations of harmful compounds, 5,6 electric devices, 7,8 optical materials, [9][10][11] ion conducting materials, 12,13 bio-and medical materials, 14,15 nanomaterials, [16][17][18][19] and environmental materials. 3,4,7,8 Recently, we reported the preparation of the DNA-inorganic hybrid film 20 and DNA-immobilized glass beads 21 by mixing the double-stranded DNA and silane coupling reagents.…”
