Double‐blind trial of tetracycline in recurrent aphthous ulceration

Abstract: A double-blind trial of a tetracycline suspension was carried out in 25 patients with recurrent aphthous oral ulcerations. A combined topical-systemic treatment procedure was used. Both the placebo and tetracycline groups experienced reductions in ulcer incidence during the treatment period, whereas only the tetracycline group showed significant reductions in ulcer duration, size, and pain. Tetracycline treatment apparently alters the severity of the ulcer, but does not appear to affect those factors responsib… Show more

“…Amlexanox is an anti-inflammatory, anti-allergic and immunomodulatory (not currently available in the USA). Two reasonably sized double-blind trials (100-200 RAS patients) showed that Amlexanox oral adhesive tablets applied 4 times a day for 5 days were effective at promoting healing and reducing pain [57,58] Topical tetracycline mouthwash has been used alone or in combination with liquid anti-fungals or topical steroids, especially in the treatment of Herpetiform RAS and remains the treatment of choice in this type of RAS which appears largely resistant to steroids [59]. In major and minor RAS, topical tetracycline or minocycline mouthwashes as a local anti-bacterial can be expected to reduce the severity of the ulcerations and pain but not prevent recurrences [60,61].…”

“…The treatment is mainly based on the suppression of inflammatory attacks of the disease using immunomodulatory and immunosuppressive agents. Based on the mainly controlled studies and personal experience in clinical practice and basic research in this field, a stepwise, Amlexanox versus placebo Decrease in the pain severity and healing duration [78][79][80] 5-aminosalicylic acid versus placebo Decreases the healing duration 81 Tetracycline versus placebo Reductions in ulcer duration, size and pain 72,73 Triclosan versus placebo Reduces the number of aphthous ulcers 74 Camel thorn distillate versus placebo Reductions in ulcer size and the pain severity …”

“…Tetracycline mouthwash (250 mg capsules dissolved in 5 mL of water or flavored syrup and held in the mouth for about 2 minutes before swallowing four times daily) decrease pain severity and duration of OUs. A double-blind trial of tetracycline suspension showed significant reductions in ulcer duration, size, and pain in RAS patients [23]. A recent study [24] assessed 0.2 percent minocycline and 0.25 percent tetracycline aqueous solution mouthwash in patients with RAS in a clinical randomized crossover trial.…”
