Dorsoventral Differences in Intrinsic Properties in Developing CA1 Pyramidal Cells

Abstract: The dorso-ventral and developmental gradients of entorhinal layer II cell grid properties correlate with their resonance properties and with their hyperpolarization-activated cyclic nucleotide-gated ion channel current (HCN, Ih) characteristics. We investigated whether such correlation existed in rat hippocampal CA1 pyramidal cells, where place fields also show spatial and temporal gradients. Resonance was absent during the first postnatal week, and emerged during the second week. Resonance was stronger in dor… Show more

“…Indeed, in rats, membrane conductances near rest differ from DH to VH (i.e. Kv723, HNCN24 and GIRK25) and dendritic surface area of DH neurons is larger than that of VH ones101126. We did not observe any significant difference in the average membrane capacitance between VH and DH cells but we measured a similar specific conductance, suggesting that differences in RMP and Rin might depend on both cell dimension and ionic conductances.…”

“…Surprisingly, the dorso-ventral gradient of resonance and I h properties of CA1 pyramidal cell dendrites are reversed as compared with those found at earlier stages of development (13). The meaning and mechanisms for such changes remain to be determined.…”

“…In Vitro Electrophysiology-Hippocampal 350-m-thick dorsal (coronal) or ventral (sagittal with a 45°angle) slices were prepared from male adult Wistar Han rats, as described (13). Slices were then transferred to a holding chamber at room temperature in normal artificial cerebrospinal fluid.…”

“…More generally, while most studies to date focused on the dorsal hippocampus, substantial cognitive (e.g. fear vs. declarative learning), connectivity (Ropireddy and Ascoli, 2011; Cardoso-Cruz et al, 2014; Krook-Magnuson et al, 2012; Lee et al, 2014), and biophysical (Graves et al, 2012; Malik et al, 2016; Marcelin et al, 2012b; Marcelin et al, 2012a) differences exist in the septotemporal directions towards the posterior and ventral regions. Given the explicit anatomical mapping of the ISH intensity and density values across the rostro-caudal extent in the ABA, the present study could in principle be expanded in the future to account for these gradients, possibly refining the neuron type classification of Hippocampome.org.…”
