Does Bromelain-Cisplatin Combination Afford In-Vitro Synergistic Anticancer Effects on Human Prostatic Carcinoma Cell Line, PC3?

Abstract: Background: Bromelain enhances anticancer impacts to chemotherapeutic agents. The question as to whether bromelain does promote in-vitro cytotoxic and proapoptotic effects of cisplatin on human prostatic carcinoma PC3 cell line was investigated. Materials and Methods: PC3 (human prostatic carcinoma) cells were treated either single or in combination with bromelain and/or cisplatin. MTT, clonogenic assay, flow cytometry and real-time quantitative polymerase chain reaction were used to investigate cell viability… Show more

“…Cisplatin-bromelain showed a synergistic antitumor effect in vitro by inhibiting colony formation, in addition to increased p53 proapoptotic gene expression. Notably, the use of bromelain significantly allowed reduction in the required dose of cisplatin [166]. Similar synergistic effects were observed when using the bromelain-cisplatin combination against malignant peritoneal mesothelioma cells.…”

“…The synergistic effect of bromelain with another chemotherapeutic agent-cisplatinhas been confirmed in a number of studies using a breast cancer model [163][164][165], human prostatic [166], and gastric [165] carcinoma, and peritoneal mesothelioma cells [153,167]. Synergistic enhancement of the antitumor effect was observed in MD-MB-231 and MCF7 cells treated with the combination of bromelain and cisplatin compared to bromelain or cisplatin single treatment.…”

“…Dox is an anthracycline class medication derived from the Streptomyces peucetius bacterium that uses as antibiotic and anti-tumor agent (7). In the meantime, medicinal plants and their derivatives have been considered due to their fewer side effects, cheapness, readily availability and their antitoxic effects on healthy cells (8)(9)(10). One of the phytochemicals whose positive effects have been proven in various diseases is berberine (Ber).…”

“…Combination therapy is used in multifactorial conditions to achieve more efficient treatment. It targets cancer's key pathways explicitly with a synergistic effect and reduces the dose of chemotherapy (Bozic et al 2013;Korkut et al 2015;Amini Chermahini et al 2020). This method potentially decreases drug resistance while retaining and strengthening the therapeutic potential of anti-cancer reagents such as trapping cells in the cell cycle, inducing apoptosis and limiting the cancer stem cells population, tumor growth, angiogenesis, and metastatic ability (Foucquier and Guedj 2015).…”
