Does 5-Hydroxytryptophan Cause Acute and Chronic Toxic Perturbations in Rats?

Abstract: In 1989, an epidemic of eosinophilia-myalgia syndrome (EMS) occurred in the United States that was attributed to contaminated l-tryptophan (LT). Features of tryptophan-induced EMS included debilitating myalgia and marked peripheral eosinophilia. Although the contaminant(s) was found only in the product produced by a single manufacturer (Showa Denko), all LT was withdrawn from the market and replaced by 5 hydroxytryptophan (5HTP). The belief was that the latter should not contain the implicated contaminant(s), … Show more

“…The findings of this present study are also comparable to those of Preuss et al ., who described no significant evidence of eosinophilia–myalgia syndrome – a rare condition that affects muscles, skin, and lungs in rats receiving high-dose 5-HTP for 1 year. [ 24 ] Despite the fact that 5-HTP has been used for the past 20 years in countries all over the world, aside from one unsolved case involving a Canadian woman, no confirmed cases of toxicity have emerged. [ 6 ] Taken together, the current study results with the lack of toxicity provide further support for the safety of administration of 5-HTP + AET.…”

“…Similar, in rats, a 1-year toxicology study found no effects of oral high-dose 5-HTP (875 mg/kg/day, via the drinking water) on cardiovascular, hepatic, renal, hematological, body weight gain, organ histology, and organ weight parameters [74]. In humans, common adverse events seen with oral 5-HTP are mild to moderate, and gastrointestinal, e.g.…”
