DNA sequence, physics, and promoter function: Analysis of high-throughput data On T7 promoter variants activity

Abstract: RNA polymerase/promoter recognition represents a basic problem of molecular biology. Decades-long efforts were made in the area, and yet certain challenges persist. The usage of certain most suitable model subjects is pivotal for the research. System of T7 bacteriophage RNA-polymerase/T7 native promoter represents an exceptional example for the purpose. Moreover, it has been studied the most and successfully applied to aims of biotechnology and bioengineering. Both structural simplicity and high specificity of… Show more

“…The list of predicted p moters includes the promoters anticipating the so-called 'early region' genes in the beg ning of the genome, the promoters of DNA and RNA polymerases' genes, the promot anticipating the blocks of structural and lysis genes. The arrangement of predicted p moters is close to that one of the model phage T7 [39]. The genetic map of phage Aristop anes is shown in Figure 3.…”

“…The recognition region (positions − 17 to -5) of the T7 native promoter includes the AT-rich recognition loop (positions − 17 to -13) and the speci city loop (positions − 12 to -5). These provide a sequence-speci c recognition by the bacteriophage T7 RNA-polymerase (RNAP) [55]. The T7 RNAP can recognize the sequences closely related to the T7 native promoter [56].…”
