DNA content of Ovis musimon spermatozoa

Abstract: To our knowledge, the value of the haploid DNA content (C-value) of Ovis musimon (mouflon) has not been previously published. Therefore, the aim of the present work was to determine the C-value and the nuclear area of O. musimon sperm cells and compare both parameters with those of Ovis aries. Feulgen reaction, which is specific and stoichiometric for DNA, was carried out on semen smears. The C-value and sperm nuclear area were determined using microspectrophotometry and Gallus domesticus erythrocytes as stand… Show more

“…The haploid DNA content (C‐DNA) per sperm nucleus expressed in picograms was calculated using G. domesticus erythrocytes as a standard (C‐value = 1.25 pg, Gregory, ) and according to Spirito, Campi, Boquet, Fernández, and Ferrari () and Ferrari et al. (). The scanning method allowed obtaining densitometric parameters related to the DNA distribution and condensation in each sperm nucleus: point absorbance (measure of the absorbance of each of the points), mean absorbance (mean obtained from the absorbance of all the points), maximal absorbance (absorbance of the point with the largest absorbance) and maximal absorbance/mean absorbance (relation between maximal and mean absorbance).…”

“…DNA content was measured in a pool of 210 morphologically normal sperm nuclei obtained from seven llama males. The amount of DNA–Feulgen complex, expressed in arbitrary units as mean absorbance per area, was obtained using the scanning method in a Cytoscan Zeiss Universal microscope (UMSP 30, Oberkochen, Germany) at a wavelength of 560 nm according to Ferrari, Spirito, Giuliano, and Cisale ().…”
