Since M. sinensis Anderss., M. sacchariflorus (Maxim.) Hack. and M. ×giganteus J.M.Greef & Deuter ex Hodk. and Renvoize have considerably the highest potential for biomass production among Miscanthus Anderss. species, there is an urgent need to broaden the knowledge about cytological characteristics required for their improvement. In this study our objectives were to assess the genome size variation among eighteen Miscanthus accessions, as well as estimation of the monoploid genome size (2C and Cx) of the M. sinensis cultivars, which have not been analyzed yet. The characterization of three Miscanthus species was performed with the use of flow cytometry and analysis of the stomatal length. The triploid (2n = 3x = 57) M. sinensis 'Goliath' and M. ×gigan-teus clones possessed the highest 2C DNA content (8.34 pg and 7.43 pg, respectively). The intermediate 2C-values were found in the nuclei of the diploid (2n = 2x = 38) M. sinensis accessions (5.52-5.72 pg), whereas they were the lowest in the diploid (2n = 2x = 38) M. sacchariflorus ecotypes (4.58-4.59 pg). The presented study revealed interspecific variation of nuclear DNA content (P<0.01) and therefore allowed for recognition of particular taxa, inter-and intraspecific hybrids and prediction of potential parental components. Moreover, intraspecific genome size variation (P<0.01) was observed in M. sinensis cultivars at 3.62%. The values of the stomatal size obtained for the triploid M. ×giganteus 'Great Britain' (mean 30.70 μm) or 'Canada' (mean 29.67 μm) and diploid M. sinensis 'Graziella' (mean 29.96 μm) did not differ significantly, therefore this parameter is not recommended for ploidy estimation.K Ke ey yw wo or rd ds s: : Flow cytometry, intraspecific variation, Miscanthus, nuclear DNA content, stomatal size

Interspecific hybrids between Miscanthus sacchariflorus and Miscanthus sinensis, such as Miscanthus × giganteus, are promising biomass crops for temperate regions. A set of DNA markers to distinguish both species would be a powerful tool for interspecific hybrid breeding. Out of 85 intron‐flanking primer pairs developed using genomic information from sugarcane (Saccharum hybrid) and Sorghum bicolor, seven were selected that showed polymorphisms between M. sacchariflorus and M. sinensis. Four simple sequence repeat primer pairs were also screened that showed polymorphisms between both species from 44 previously developed primer pairs. Of the polymerase chain reaction (PCR) products generated by the 11 primer pairs, nine amplified fragments were specific to M. sacchariflorus and two were specific to M. sinensis, based on 83 accessions mainly from Japan but also including Chinese and Korean ones. Fragments specific to two species were detected in three known hybrid plants. These results indicate that combinations of these PCR fragments could be good markers for identifying hybrids of M. sacchariflorus and M. sinensis obtained from breeding programs or from natural crosses between wild sympatric populations.

Analyzing the ploidy levels of plants is important for identifying species, selecting parental lines, identifying the relationships between species, and determining evolutionary patterns. The genus Chrysanthemum is widely distributed throughout the world and exhibits different ploidy levels. We used flow cytometry to analyze the ploidy levels of nine species of Chrysanthemum L. collected from different regions and geographical locations in China. Three diploids from Henan and Wuhan provinces corresponded to Chrysanthemum lavandulifolium and two species of C. nankingense, while three tetraploids from various regions corresponded to C. indicum and two species of C. chanetii. Two hexaploids corresponding to C. vestitum were collected at Funiu mountain (Henan province), and C. zawadskii was collected at Huangshan mountain (Anhui province). We found that OTTO extraction buffer was suitable for extracting nuclei from most species, apart from C. zawadskii. Flow cytometry proved to be a simple, rapid, and highly accurate method for identifying ploidy levels in Chrysanthemum species.
