Diverse Functions of Il-2, Il-15, and Il-7 in Lymphoid Homeostasis

Abstract: IL-2, IL-15, and IL-7 are cytokines that are critical for regulating lymphoid homeostasis. These cytokines stimulate similar responses from lymphocytes in vitro, but play markedly divergent roles in lymphoid biology in vivo. Their distinct physiological functions can be ascribed to distinct signaling pathways initiated by proprietary cytokine receptor chains, differential expression patterns of the cytokines or their receptor chains, and/or signals occurring in distinct physiological contexts. Recently, the di… Show more

“…Many studies have demonstrated that anti-tumor immunity and the ability to break tolerance could be enhanced by co-delivering additional immunomodulator genes, such as interleukin-2, interleukin-12, interleukin-15, CD70 and other activators, to DCs. 5,[57][58][59][60] We are currently conducting experiments to evaluate the anti-tumor immunity generated by the co-delivery of a tumor antigen and an immuomodulator using this DC-directed lentivector system.…”

“…Activated OT-II cells expressed the subunits a (CD25) and b (CD122) of the IL-2 receptor at the time of adoptive transfer, whereas naive OT-II did not. The source of the IL-2 is clearly the TT-specific CD4 1 T cells [22], where production is usually accompanied by upregulation of the subunits a and b of IL-2R [22] and would be followed by autocrine and/or paracrine consumption of the cytokine [22]. IL-2 is therefore a strong candidate cytokine for driving proliferation of the injected OT-II cells.…”

“…IL-2 is a proinflammatory cytokine and one of the most essential cytokines for regulating activation and proliferation of t-cells [20]. The TGF-β1 protein family is involved in muscle growth and atrophy [21] and has been implicated in both the inhibition and activation of t-cells [22,23].…”
