Diterpenoid alkaloids from some Turkish Consolida species and their antiviral activities

Abstract: Five diterpenoid-derivative alkaloids, lycoctonine (1), 18-O-methyllycoctonine (2), delcosine (3), 14-acetyldelcosine (4), and 14-acetylbrowniine (5), were screened for their antibacterial, antifungal, and antiviral activities. Both DNA virus Herpes simplex (HSV) and RNA virus Parainfluenza (PI-3) were employed for antiviral assessment of the compounds using MadinDarby bovine kidney and Vero cell lines. Their maximum non-toxic concentrations (MNTC) and cytopathogenic effects (CPE) were determined using acyclov… Show more

“…Standard antibacterial powders of ampicilline (AMP, Fako), ofloxacine (OFX, Hoechst Marion Roussel), and also standard antifungal powders of ketoconazole (KET, Bilim) and fluconazole (FLU, Pfizer) were obtained from their respective manufacturers and dissolved in phosphate buffer solution (AMP, pH: 8.0, 0.1 mol/L), DMSO (KET), in water (FLU). The stock solutions of the agents were prepared in media according to the NCCLS recommendations as referred in our previous studies [13][14][15][16]. Acinetobacter baumannii (RSKK 02026), Staphylococcus aureus (ATCC 25923), and Bacillus subtilis (ATCC 6633) for antibacterial activity, as well as a standard strain of Candida albicans (ATCC 10231) were employed.…”

“…Strains of HSV and PIV titers were calculated by the Frey and Liess method as TCID 50 [18]. The assay was performed as reported previously and maximum cytopathogenic effect (CPE) concentrations, as the indicator of antiviral activities of the extracts, were determined [13][14][15][16].…”

“…Diterpenoid alkaloids are a very important family of natural products that feature structural complexity and various bioactivities, such as anti-inflammatory [3][4], analgesic, antiarrhythmic, anti-epileptiform, anticancer, antiparasite and anesthetic activities [5][6]. Most natural diterpenoid alkaloids were isolated from the genera Aconitum [7], Consolida [8] and Delphinium(Ranunculaceae) [9] and the genus Spiraea (Rosaceae) [10]. As part of our research project to explore more bioactive lead compounds from the medicinal herbs in the Qinba mountains of China, the chemical constituents and pharmacological studies of Aconitum sinomontanum were studied, and one new C18-diterpenoid alkaloid sinomontanine I (1), along with four known diterpenoid alkaloids, delcosine (2) [11], lepenine (3) [12], napelline(4) [13], and kirinine B (5) [12] were isolated (Figure 1).…”
