Distributions of high-density lipoprotein particle components in human follicular fluid and sera and their associations with embryo morphology parameters during IVF

Abstract: In this pilot study, our data suggests that HDL and its component proteins within FF may play protective roles in the health of the human oocyte and subsequent early embryo development. We describe for the first time the activities of PON1 and PON3 in FF. We suspect that PON3 activity may be locally generated due to higher activities in FF compared with serum.

“…11,38 To our knowledge, the first study investigated PON activity in FF was conducted by Browne et al and they have found that PON1-arylesterase activity in FF was a positive predictor for Day 3 embryo cell number. 10 Contrary to that study, we found similar FF levels of PON1 and arylesterase activity in women with PCOS and in controls. It is thought that proven conditions including increased oxidative stress in women with PCOS is not the cause of the infertility in those women, however; in our study, differently from other studies, the oxidative molecules have been investigated in follicle fluid representing the biological window of ovary.…”

“…9 The effect of oxidative stress on etiology of PCOS and effect of PON1 in women with PCOS are still debate; there are a few studies with conflicting results about relationship between PON1 and PCOS. [10][11][12][13] Therefore we aimed to compare the follicular fluid level of PON and ARES activity in women with PCOS and those with Male factor infertility (MFI) who underwent assisted reproductive technique (ART) using intra-cytoplasmic sperm injection (ICSI).…”

“…Thus, the structure, size, and antioxidant activity of HDL are dynamic. We previously conducted a pilot study of HDL particle components, including lipids, ApoA-1, and paraoxonase 1 (PON1) activities measured in FF and human serum, and their associations with embryo quality on 60 women undergoing in vitro fertilization (IVF) [21]. In that study, we identified FF ApoA-1 and HDL-cholesterol (HDL-C) as predictors of embryo fragmentation, suggesting protective associations for embryo quality.…”

“…Arylesterase and paraoxonase activities were also analyzed using the Cobas Fara II as described elsewhere [22]. FF HDL fractions were prepared by selective precipitation to remove trace amounts of apolipoprotein B containing low density lipoproteins [17].…”

“…Given its potent antioxidant activity [12][13][14], and critical role in delivering cholesterol for steroid hormone synthesis [15,16], HDL has strong potential as a biomarker of the follicular microenvironment. Studies also identified HDL particle components measured in FF, including cholesterol, ApoA-1 [17], γ-tocopherol, and β-cryptoxanthin [18], as important predictors of embryo quality during in vitro fertilization (IVF). Higher FF apolipoprotein A-1 (ApoA-1) levels were recently associated with poorer embryo quality per oocyte, in women undergoing IVF [19].…”
