Aims This study attempted to demonstrate the vertical shift in bacterial, archaeal and fungal communities along the vadose zone–aquifer sediments and their respective responses to environmental factors. Methods and Results We collected samples from the vadose zone and three aquifer sediments along a 42·5 m bore of a typical agricultural land. The results showed that the bacterial community shifted greatly with depth. The classes of Actinobacteria (19·5%) and NC10 (11·0%) were abundant in the vadose zone while Alphaproteobacteria (22·3%) and Gammaproteobacteria (20·1%) were enriched in the aquifer. Archaeal and fungal communities were relatively more homogeneous with no significant trend as a function of depth. Process analyses further indicated that selection dominated in the bacterial community, whereas stochastic processes governed archaeal and fungal communities. Moreover environment–bacteria interaction analysis showed that metal(loid)s, especially alkali metal, had a closer correlation with the bacterial community than physicochemical variables. Conclusions Depth strongly affected bacterial rather than archaeal and fungal communities. Metal(loid)s prevailed over physicochemical variables in shaping the bacterial community in the vadose zone–aquifer continuum. Significance and Impact of the Study Our study provides a new perspective on the structure of microbial communities from the vadose zone to the deep aquifers.

Variations in dissolved organic matter (DOM) quality has far-reaching implications that affect, e.g., aquatic productivity, food web structures, trace element and pollutant transport. In this study, a total of 186 water samples were collected at 62 sites (three points in time within one year) in Nansi Lake. UV-Vis spectra, Synchronous fluorescence (SF) spectra, the excitation-emission matrix and parallel factor analysis (EEM-PARAFAC) were applied to indicate the source and quality of DOM. Water transferring of the eastern route of China’s South-to-North Water Diversion Project had a great influence on the water level of Nansi Lake. Results of SF spectra, EEM-PARAFAC and principal component analysis (PCA) suggested that protein-like substances played a more important role in DOM properties in April and July than October. This result is related to a high fluorescence intensity occurred in April (Fmax=0.72±0.03 in the upper lake and 1.84±0.13 in the lower lake) and July (Fmax=1.10±0.05 in the upper lake and 1.49±0.04 in the lower lake), which might be caused by water transferring from other lakes to Nansi Lake, death and decomposition of submerged plants. At the same time, relatively good correlations were found between humic-like substances, DOC and a254 in April, July and October, which indicated the important contribution of humic-like substances to Nansi Lake. With the completion of the water diversion, the ratio of the fluorescence intensity of component to the total fluorescence intensity (%Fmax) suggested that the proportion of humic-like substances started to increase. And when it came to October, humic-like substances become the main substance in DOM collected from Nansi Lake (%Fmax=66.56%±0.58% in the upper lake and 61.98%±0.99% in the lower lake). Moreover, among the two areas in Nansi Lake, the upper lake always had a higher degree of humification (HIX=2.23±0.06, 2.38±0.11 and 3.10±0.05 in April, July and October, respectively) than the lower lake (HIX=1.06±0.05, 1.68±0.05 and 2.62±0.08 in April, July and October, respectively), which implied extraneous contaminants might have a more important impact on DOM properties in the upper lake.

The Pietra del Pertusillo freshwater reservoir is a major artificial lake of environmental, biological, and ecological importance located in the Basilicata region, southern Italy. The reservoir arch-gravity dam was completed in 1963 for producing hydroelectric energy and providing water for human use, and nearby there are potential sources of anthropogenic pollution such as urban and industrial activities. For the first time, the minero-chemistry of the lake and fluvio-lacustrine sediments of the reservoir have been evaluated to assess the environmental quality. Moreover, the composition of fluvial sediments derived from the peri-lacual zone of the reservoir and of local outcropping bedrock were also studied to understand the factors affecting the behavior of elements in the freshwater reservoir, with particular attention paid to heavy metals. In Italy, specific regulatory values concerning the element threshold concentration for lake and river sediments do not exist, and for this reason, soil threshold values are considered the standard for sediments of internal waters. The evaluation of the environmental quality of reservoir sediments has been performed using enrichment factors obtained with respect to the average composition of a reconstructed local upper continental crust. We suggest this method as an innovative standard in similar conditions worldwide. In the studied reservoir sediments, the trace elements that may be of some environmental concern are Cr, Cu, Zn, As, and Pb although, at this stage, the distribution of these elements appears to be mostly driven by geogenic processes. However, within the frame of the assessment and the preservation of the quality of aquatic environments, particular attention has to be paid to As (which shows median value of 10 ppm, reaching a maximum value of 26 ppm in Quaternary sediments), constantly enriched in the lacustrine samples and especially in the fine-grained fraction (median = 8.5 ppm).
