Distribution of ABCB1 Polymorphisms Among Brazilians: Impact of Population Admixture

Abstract: Self-identification according to the racial/color categories proposed by the Brazilian Census is insufficient to properly control for population stratification in pharmacogenomic studies of ABCB1.

“…[35,36] No significant differences between C/T and G/T were detected in our study, in contradiction with other studies previously published on Asian populations. [37,38] A strong association for C1236T and G2677T linkage disequilibrium was observed in a Macedonian study. [32] When we analysed the linkage between these two SNPs the most frequent genotype we found was CT/GT, followed by CC/GG and TT/TT, but no statistical differences were detected between the study groups or associated with drug-resistant epilepsy.…”

“…Methods that determine genetic ancestry using patient genotype data reflect differences in allele frequencies significantly more accurately. 4,16,34 Preferably, these methods should be fast and easy to use even on large datasets without the need for Figure 3 Scatter plot of PC1 and PC2 of GATC samples with self-reported geographic origin from Canada. PCA shows that the genetic structure of most of these individuals is similar to Europeans.…”

“…This strategy has been applied previously [21,23,24] and is consistent with alleles *3, *6 and *7 being functionally inactive [27]. ABCB1 polymorphisms were genotyped by the single base extension/termination method using the SNaPshot ® multiplex system from Applied Biosystems as previously described [28]. Exonic fragments containing the SNPs ABCB1 1236C>T, 2677G>T/A and 3435C>T were amplified in a multiplex-PCR, using primers 5´TATTCG-A AGAGTGGGCACA A3´ (forward) and 5´CTGATCAC CGCAG GGTCTA3´ (reverse), 5´AGCAA ATC TTGGGACA GGAA3´ (forward) and 5´AAGATTGCT TTGAGGAATGGTT3´ (re verse ), 5´G AG CCC AT CC TGT T T-GACTG3´ (forward) and 5´CATGCTCCCA-GGCTGTTTAT3´ (reverse), respectively.…”
