Distinct roles of bone morphogenetic proteins in osteogenic differentiation of mesenchymal stem cells

Abstract: Efficacious bone regeneration could revolutionize the clinical management of many bone and musculoskeletal disorders. Bone morphogenetic proteins (BMPs) can regulate the differentiation of mesenchymal stem cells into cartilage, bone, tendon/ligament, and fat lineages. Early data documented the osteogenic potential of rhBMP2 and rhBMP7/OP-1. However, prior to this work that summarized several of our recent studies, no comprehensive analysis had been undertaken to characterize relative osteogenic activity of all… Show more

“…9,10,40 The cell cultures used in our study constitute a mixed population of different cell types and stages of differentiation to mimic in vivo cell populations. Nevertheless, bone formation is a complex process that may occur through endochondral or intramembraneous ossification preceded by bone resorption.…”

“…Few studies have addressed the combined effects of BMPs on osteogenesis, mainly focusing on BMP-2 and BMP-7. 9,10 We showed that BMP-2, BMP-5, and BMP-6 alone have distinct and diverse effects on osteoclasts and osteoblasts in vitro. 8 The aim of the present study was to investigate the effects of the simultaneous use of BMP-2, BMP-5, and BMP-6 on osteoblast and osteoclast generation to test the hypothesis that BMPs in combination exert synergistic effects on processes of bone regeneration.…”

“…2,3 Mesenchymal stem cells (MSCs) are adherent bone marrow stromal cells that can differentiate into osteogenic, chondrogenic, adipogenic, and myogenic lineages. [17][18][19][20][21] Osteoblastic differentiation of MSCs is an essential process of bone formation, [17][18][19][20] and is tightly regulated. 20,21 BMPs play an important role in osteoblast differentiation.…”

“…(1,2) Several BMPs play an especially critical role in differentiation of precursor cells to induce bone formation. (3) Of the BMPs, BMP-2 is the most well studied inducer of osteoblast differentiation and bone formation. (4) BMP-2 is approved for clinical application to induce new bone formation in spine and severe fracture patients, but the amount of recombinant protein required makes its use prohibitively expensive.…”
