Low-density lipoprotein (LDL) cholesterol, high-density lipoprotein (HDL) cholesterol, triglycerides, and total cholesterol are heritable, modifiable, risk factors for coronary artery disease. To identify new loci and refine known loci influencing these lipids, we examined 188,578 individuals using genome-wide and custom genotyping arrays. We identify and annotate 157 loci associated with lipid levels at P < 5×10−8, including 62 loci not previously associated with lipid levels in humans. Using dense genotyping in individuals of European, East Asian, South Asian, and African ancestry, we narrow association signals in 12 loci. We find that loci associated with blood lipids are often associated with cardiovascular and metabolic traits including coronary artery disease, type 2 diabetes, blood pressure, waist-hip ratio, and body mass index. Our results illustrate the value of genetic data from individuals of diverse ancestries and provide insights into biological mechanisms regulating blood lipids to guide future genetic, biological, and therapeutic research.
The recent discovery of superconductivity in iron arsenide compounds RFeAsO (R= rare earth) or AFe 2 As 2[1~8] (A= alkaline earth) attracts great attention due to the unexpected high T c in the system containing ferromagnetic elements like Fe. Similar to high T c cuprates, the superconductivity in iron arsenide is related to a layered structure. Searching for new superconductors with [FeAs] layer but of simpler structure will be of scientific significance either to build up new multilayered superconductors that may reach higher T c or to study the mysterious underlined superconducting mechanism in iron arsenide compounds. Here we report that a new superconducting iron arsenide system LiFeAs was found. The compound crystallizes into structure containing [FeAs] conducting layer that is interlaced with Li charge reservoir. Superconductivity was observed with T c up to 18 K in the compounds.
The groundwork of intellectual capital (IC) management, measuring IC, attracts much attention from academics and practitioners. The purpose of this paper is to design a measurement model and a qualitative index system of IC, so as to provide a good tool for enterprises to manage their IC. Based on a review of several IC measurement models proposed by western researchers, IC is classiWed into human capital, structural capital, innovation capital and customer capital, and thereupon a qualitative index system for the above four IC elements is designed through an analysis of their contents. Through an empirical study, it is found that there is a signiWcant relationship between the scores of the four IC elements of a company and its business performance, which proves the validity and rationality of the IC measurement model and the qualitative index system. In the meantime, the empirical study further proves that there is a remarkable relationship between the four IC elements. Therefore enterprises must manage and improve their IC from an integrative perspective.
Ribosome profiling has revealed pervasive but largely uncharacterized translation outside of canonical coding sequences (CDSs). In this work, we exploit a systematic CRISPR-based screening strategy to identify hundreds of noncanonical CDSs that are essential for cellular growth and whose disruption elicits specific, robust transcriptomic and phenotypic changes in human cells. Functional characterization of the encoded microproteins reveals distinct cellular localizations, specific protein binding partners, and hundreds of microproteins that are presented by the human leukocyte antigen system. We find multiple microproteins encoded in upstream open reading frames, which form stable complexes with the main, canonical protein encoded on the same messenger RNA, thereby revealing the use of functional bicistronic operons in mammals. Together, our results point to a family of functional human microproteins that play critical and diverse cellular roles.
To investigate molecular mechanisms controlling islet vascularization and revascularization after transplantation, we examined pancreatic expression of three families of angiogenic factors and their receptors in differentiating endocrine cells and adult islets. Using intravital lectin labeling, we demonstrated that development of islet microvasculature and establishment of islet blood flow occur concomitantly with islet morphogenesis. Our genetic data indicate that vascular endothelial growth factor (VEGF)-A is a major regulator of islet vascularization and revascularization of transplanted islets. In spite of normal pancreatic insulin content and -cell mass, mice with -cell-reduced VEGF-A expression had impaired glucose-stimulated insulin secretion. By vascular or diffusion delivery of -cell secretagogues to islets, we showed that reduced insulin output is not a result of -cell dysfunction but rather caused by vascular alterations in islets. Taken together, our data indicate that the microvasculature plays an integral role in islet function. Factors modulating VEGF-A expression may influence islet vascularity and, consequently, the amount of insulin delivered into the systemic circulation. Diabetes
Thermally conductive but electrically insulating polymer composites are highly desirable for thermal management applications because of their wide range of utilization, ease of processing, and low cost. However, the traditional approaches to thermally conductive polymer composites usually suffer from the low thermal conductivity enhancement and/or the deterioration of electrical insulating property. In this study, using cellulose nanofiber‐supported 3D interconnected boron nitride nanosheet (3D–C–BNNS) aerogels, a novel method for highly thermally conductive but electrically insulating epoxy nanocomposites is reported. The nanocomposites exhibit thermal conductivity enhancement of about 1400% at a low BNNS loading of 9.6 vol%. In addition, the epoxy nanocomposites are still highly insulating, having a volume electrical resistivity of 1015 Ω cm. The strong potential application for thermal management has been demonstrated by the surface temperature variations of the nanocomposites with time during heating and cooling.
