Distinct Functions of Gq and G11 Proteins in Coupling α1-Adrenoreceptors to Ca2+ Release and Ca2+ Entry in Rat Portal Vein Myocytes

Abstract: In this study, we identified the subunit composition of G q and G 11 proteins coupling ␣ 1 -adrenoreceptors to increase in cytoplasmic Ca 2؉ concentration ([Ca 2؉ ] i ) in rat portal vein myocytes maintained in short-term primary culture. We used intranuclear antisense oligonucleotide injection to inhibit selectively the expression of subunits of G protein. Increases in [Ca 2؉ ] i were measured in response to activation of ␣ 1 -adrenoreceptors, angiotensin AT 1 receptors, and caffeine. Antisense oligonucleotid… Show more

“…The vasoconstrictions induced by alpha-adrenergic, angiotensin-II, and endothelin-1 stimulations implicate InsP3, CaV1.2, and NAADP [32,41,54], cADPR and CaV1.2 [18,40], and InsP3 and NAADP [6,33], respectively. In elderly humans, the vasoconstriction induced by alpha-adrenergic stimulation was decreased [52], but the mechanism was not described.…”

“…Oligonucleotides were injected into the nuclei of myocytes by a manual injection system (Eppendorf, Hamburg, Germany). Intranuclear oligonucleotide injection with Femtotips II (Eppendorf) was performed as described previously (26). The myocytes were then cultured for 3-4 days in culture medium, and the glass slides were transferred into the perfusion chamber for physiological experiments.…”

“…Briefly, oligonucleotides were designed on the known cloned sequences deposited in the GenBank database with DNAstar (Lasergene software). Oligonucleotides were injected into the nuclei of myocytes by a manual injection system with femtotips II (Eppendorf) as previously described (Macrez-Lepretre et al, 1997). Myocytes were then cultured for 2 to 4 days in culture medium.…”
