[9] However, LFAs are often limited to qualitative results readout by providing "yes" or "no" signal, which is insufficient in some application scenarios, e.g., a clinical action will be made in clinics according to the accurate amount of targets. Recently, the emerging µPADs have gained considerable interests due to their excellent capability for outputting quantitative results. [10] Nowadays, quantitative paper-based assays are mainly based on electrochemical detection, [11] electrochemiluminescence detection, [12] and fluorescent detection. [13,14] Although sensitive and quantitative detection can be obtained, special equipment for quantification of detection signal is usually involved. For instance, electrochemical workstation is needed for performing electrochemical detection on paper, while a special light-emitting device is indispensable for fluorescent testing. For addressing these limitations, some handheld equipments, such as glucose meter, [15] multimeter, [16] and smartphone, [17,18] have been introduced as readers for outputting quantitative measurements with improved portability. However, the accessibility and cost of these handheld equipments may still be a big challenge for their application in resource-poor areas, where the quantitative detection in an equipment-free manner is preferred. Hence, the exploitation of equipment-free paper-based analytical devices shows broad practical application prospects in POCT field.Colorimetric detection holds great potential for developing equipment-free detection methods through direct readout by naked eye. [19] Conventionally, the color intensity analysis has been widely used to realize quantification of analytes, whereas a camera is needed to capture images and a computer-based software for image processing. [20] Hence, these quantitative methods based on color intensity analysis are unsatisfactory for exploiting equipment-free quantitative strategies. Fortunately, thanks to the novel signal transformation design in paper-based colorimetric detection, different types of equipment-free quantitative paper devices have already been proposed, where the detection signal has been transformed into the measurement of color change distance, [21] the counting of colored detection zones, [22] the measurement of testing time, [23] and the reading of displayed text. Detection of biomarkers is essential for diagnosis and prognosis of diseases for healthcare teams to provide timely appropriate treatments. Reliable, inexpensive, and sensitive devices are desirable to serve this purpose. Paper‐based analytical devices (PADs) have gained enormous attention during the past decade as point‐of‐care devices for biomarker detection due to their simplicity, portability, and biocompatibility. Importantly, the devices highly comply with the WHO “ASSURED” concept (i.e. Affordable, Sensitive, Specific, User‐friendly, Rapid and Robust, Equipment free, and Deliverable to end‐users). Thus, PADs could be sustainably alternative tools for biomarker detection, especially in resources‐constraint areas where budget, skillful operators, and health infrastructure are limited. First, this review introduces overviews of biomarkers, their detection and brief history of PADs. Second, description of common fabrication and detection techniques in PADs are provided. Then, it highlights recent state of the art technologies and applications of PADs for various biomarker detection published during 2018 to May 2019. Applications for tumor marker, cardiac and coronary heath disease, and metabolic disorder biomarker detection are tabulated and selected examples are described in details. Finally, it discusses the current challenges, advances, and novel applications of PADs. The review would be valuable for healthcare workers using PADs as cost‐effective point‐of‐care devices for biomarker detection and for researchers working on future developments of the devices.

Immunocytological technologies, molecular technologies, and functional assays are widely used for detecting circulating tumor cells (CTCs) after enrichment from patients' blood sample. Unfortunately, accessibility to these technologies is limited due to the need for sophisticated instrumentation and skilled operators. Portable microfluidic devices have become attractive tools for expanding the access and efficiency of detection beyond hospitals to sites near the patient. Herein, a volumetric bar chart chip (V‐Chip) is developed as a portable platform for CTC detection. The target CTCs are labeled with aptamer‐conjugated nanoparticles (ACNPs) and analyzed by V‐Chip through quantifying the byproduct (oxygen) of the catalytic reaction between ACNPs and hydrogen peroxide, which results in the movement of an ink bar to a concentration‐dependent distance for visual quantitative readout. Thus, the CTC number is decoded into visually quantifiable information and a linear correlation can be found between the distance moved by the ink and number of cells in the sample. This method is sensitive enough that a single cell can be detected. Furthermore, the clinical capabilities of this system are demonstrated for quantitative CTC detection in the presence of a high leukocyte background. This portable detection method shows great potential for quantification of rare cells with single‐cell sensitivity for various applications.
