“…The extracellular concentration of these ions in large part determines the intracellular concentration (McGrady & Nelson, 1973). The cardiac glycoside ouabain, an inhibitor of cardiac Na+-K+ ATPase (Brody, 1974), has been shown to bind to Na+-K+-activated ATPase sites in the midpiece-tail of bull spermatozoa (O'Donnell & Ellory, 1970). As part of our efforts to investigate the permeability properties of mature and developing spermatozoa, the present study of freshly ejaculated bull spermatozoa considered the effect of ouabain on membrane potential, frequency of the flagellar beat and maximum amplitude of the flagellar wave in order to (a) determine the contribution of ouabainsensitive Na+-K+ transport to membrane potential in bull spermatozoa, and (b) clarify the effects of ouabain on specific characteristics of motility.…”

“…This inhibition accounts for its toxic and lethal effects, considering the widespread presence of this pump in all animal cells [ 6 ]. However, when administered at controlled doses, ouabain induces inhibition of Na + /K + -ATPase, leading to what is known as a positive inotropic effect, which explains its properties in cardiac tissue [ 7 , 8 ].…”

“…The in vitro effect of exogenous digoxin on normal human spermatozoa progressive motility Ouabain, a digitalis related compound which is known to inhibit the Na-K ATPase pump in vitro (Brody, 1974), caused severe impairment of sperm motility and acrosome reaction (Nelson & McGrady, 1981). Chulavatnatol & Yiudepit (1976) demonstrated an increase in Na-K ATPase sensitivity to ouabain inhibition in rat sperm, when it migrates from the epididymal head to its cauda.…”
