“…Isoenzyme analysis has found similar genetic variability at the biochemical level even 234 banding, CMA/DAPI staining, heterochromatin, in morphologically uniform seedling populations (Moore & Castle, 1988). Such an approach has also been useful to distinguish zygotic from nucellar seedlings (Iglesias et al, 1974;Ashari et at., 1988). As we are concerned with the cytogenetic analysis of the genus, we are interested in identifying its chromosomal variability and in looking for a further approach to the genetic characterization of species or clones of Citrus.…”

“…Andrade-Rodriguez et al (2004) were able to classify 26% of seedlings from polyembryonic seeds of C. volkameriana as zygotics, whereas Bastianel et al (1998) (1988) used isozyme markers to identify a wide range of frequency of zygotics, 5.6 to 50.6%, among the 12 different citrus rootstock accessions screened. Studies by Ashari et al (1988) reported as low as 0% of zygotic seedlings in Cleopatra mandarin using isozymes and morphological characteristics. The percentages of zygotic progenies in various citrus hybrids have been found to depend on the seed parent used (Spiegel-Roy et al 1977), the pollen origin (Cameron and Soost 1969), and environmental influences (Moore and Castle 1988).…”

“…Various techniques have been used to distinguish nucellar from zygotic offspring including colorimetric assays (Furr and Reece 1946), infrared spectroscopy (Pieringer and Edwards 1967), enzymatic darkening of polyphenols (Esen and Soost 1974), chromatography (Weinbaum et al 1982), and isozyme pattern analysis (Ashari et al 1988; Moore and Castle 1988;Xiang and Roose 1988;Tusa et al 2002). However, none of these methods efficiently provide sufficient confirmation of the identity of true nucellar seedlings (Ruiz et al 2000;Tusa et al 2002).…”

(Ruiz et al, 2000;Tusa et al, 2002).…”
