Discrimination between protein glycoforms using lectin-functionalised gold nanoparticles as signal enhancers

Abstract: Biolayer interferometry is used to distinguish between glycoforms of the same using lectin-functional nanoparticles as both readers, and signal enhancers. The use of this is exemplified with prostate specific antigen.

“…Lectins are natural proteins that are able to recognize bound monosaccharides, oligosaccharides, and glycans attached to the proteins, thus enabling recognition of the glycoproteins. Lectins have been utilized as recognition elements or bioreceptors in the fabrication of biosensors tailored for glycoprotein determination. ,, A general requirement of impedimetric biosensors is to utilize small bioreceptors for protein detection . Lectins are generally smaller than antibodies, and thus they are ideal candidates to serve as small bioreceptors, being able to form a denser biofilm on the sensor surface to enable sensitive detection.…”
