Discovery of a trans -Dichloroethene-Respiring Dehalogenimonas Species in the 1,1,2,2-Tetrachloroethane-Dechlorinating WBC-2 Consortium

Abstract: bThe WBC-2 consortium is an organohalide-respiring anaerobic microbial enrichment culture capable of dechlorinating 1,1,2,2-tetrachloroethane (TeCA) to ethene. In the WBC-2 culture, TeCA is first transformed to trans-dichloroethene (tDCE) by dichloroelimination; tDCE is subsequently transformed to vinyl chloride (VC) and then to ethene by hydrogenolysis. Analysis of 16S rRNA gene clone libraries from culture DNA revealed sequences from three putative dechlorinating organisms belonging to Dehalococcoides, Dehal… Show more

“…This consortium contains Dehalobacter, Dehalococcoides and Dehalogenimonas strains [104]. Previous sequence data from this consortium have been limited to 16S rRNA gene clones, leaving the reductive dehalogenase complement unknown.…”

“…The West Branch Canal Creek Consortium (WBC-2) culture was derived from sediments contaminated with 1,1,2,2-tetrachloroethane (1,1,2,2-TeCA) (10). In this consortium, 1,1,2,2-TeCA dehaloelimination to trans-dichloroethene (tDCE) is carried out by Dehalobacter, tDCE dechlorination to vinyl chloride (VC) is carried out by Dehalogenimonas, and VC dechlorination to ethene is carried out by Dehalococcoides (11). On a 16S rRNA gene sequence basis, the Dehalogenimonas population from the WBC-2 culture is 96% similar to Dehalogenimonas lykanthroporepellens, while it is only 91% similar to Dehalococcoides (11).…”

The Dehalogenimonas carrying out the middle step is of particular interest because it is the first non-Dehalococcoides OHRB found to dechlorinate a lesser-chlorinated dichloroethene isomer (11).…”
