The present study details a new method for the exposure and viewing of individual microvessels located within the small intestine of rats. This procedure will selectively and consistently remove the outer muscle layers and underlying submucosa of the intestinal wall and thereby expose a variety of arterioles in their normal location within the tissue, with their normal relationship to each other undisturbed. The small intestine of the rat was initially fixed by vascular perfusion with 2.5% glutaraldehyde in 0.1 M cacodylate/HCL buffer, reinfused with heparinized whole blood, removed from the animal, and secured to a dissecting petri dish for further fixation. Subsequently, the external muscularis was dissected from the sample which exposed the submucosa. In order to remove the connective tissue elements from this layer and uncover the submucosal vasculature, the samples were first transferred to a solution of 30% potassium hydroxide for 2-5 minutes and then to a final digesting solution containing collagenase. Thereafter, the samples were routinely processed for light microscopy and for scanning (SEM) or transmission (TEM) electron microscopy. Examination of the samples revealed excellent preservation of the three-dimensional organization of the arteriolar wall with minimal membrane damage. This new technique now makes it possible to visualize the shape and position of individual smooth muscle cells along arterioles of differing size and branching order.

The combined use of scanning electron-microscopy and chemical separation of epidermis permitted the study of the irradiated and normal hair follicle surface characteristics. Irradiation reduced the length and density of the hair follicles, producing at the same time marked alterations and deformities of the individual follicles. As most of these changes were dose- and time-dependent, these structures, studied by these techniques, provide an interesting model as biological indicators of radiation dose.

SUMMARY A simplified technique has been used to examine the epidermis in five skin biopsies and in eight cultured skin specimens by scanning electron microscopy. Paraffin or cryostat sections of the tissue were cut and prepared according to routine histological methods and then given an electrically conducting coating before examination in the scanning electron microscope. The epidermis was easily identified and a particularly good view was obtained of intercellular bridges at high magnifications. Intra‐epidermal and basal layer dendritic cells were also easily located and their relationships to surrounding keratinocytes by long intercellular processes are described. The scanning electron microscopic appearance of epidermal cells in cultured explains of skin showed a strong resemblance to other cells and tissues in culture and also demonstrated the presence of a unicellular or subcellular membrane at the leading edge similar in appearance to the undulating membrane of fibroblasts in culture. It is stressed that the method is of particular use in the examination of intercellular relationships within the epidermis and in the inspection of histological material at higher magnifications than are possible by light microscopy.
