Direct Reprogramming of Fibroblasts via a Chemically Induced XEN-like State

Abstract: Direct lineage reprogramming, including with small molecules, has emerged as a promising approach for generating desired cell types. We recently found that during chemical induction of induced pluripotent stem cells (iPSCs) from mouse fibroblasts, cells pass through an extra-embryonic endoderm (XEN)-like state. Here, we show that these chemically induced XEN-like cells can also be induced to directly reprogram into functional neurons, bypassing the pluripotent state. The induced neurons possess neuron-specific… Show more

“…Transplantation of parenchymal stem cells might aid in reinstating the regenerative ability (2038), but efficient engraftment of parenchymal cells is handicapped by the prohibitive fibrotic microenvironment in diseased organs. Therefore, designing effective cell therapy strategies requires understanding how microenvironmental cues regulate parenchymal regeneration and fibrosis in the damaged lung and liver (3947).…”

“…Before using iPSCs clinically, the inserted transgenes have to be either deleted or effectively silenced after the cells have been reprogrammed. With other approaches, for example using nonintegrating vectors [104,105] , introduction of stemness proteins [106,107] , or pharmaceutically with a suitable combination of small molecules, have been developed to address this problem to a certain extent [44,108,109] . Recently, a new method to generate iPSCs with CRISPR activation through precise epigenetic remodeling of endogenous loci shed light on how targeted chromatin remodeling triggers pluripotency induction [110] .…”
