Direct Proinflammatory Effect of C-Reactive Protein on Human Endothelial Cells

Abstract: CRP induces adhesion molecule expression in human endothelial cells in the presence of serum. These findings support the hypothesis that CRP may play a direct role in promoting the inflammatory component of atherosclerosis and present a potential target for the treatment of atherosclerosis.

“…The inflammatory cell types typically found in the atheroma include monocyte-derived macrophages and lymphocytes [18]. Macrophages present in the atherogenous plaque lead to the release of mediators like cytokines and chemokines which in turn increase the plasma concentration of CRP which amplify inflammatory and procoagulant responses [19]. Several other inflammatory markers for prediction of coronary accidents in healthy men and women and in patients with stable angina, acute coronary diseases and in follow up of cardiovascular accident have been studied.…”

“…Liver is the major producer of CRP, and circulating CRP is an independent risk factor for cardiac events (19). CRP has also been shown to be produced in atherosclerotic lesions (20), and to induce adhesion molecule expression in endothelial cells (21), perhaps playing a direct role in promoting the inflammatory component of atherosclerosis. However in the Dallas Heart Study circulating CRP levels were not directly associated with atherosclerosis extent (22), similar to our own findings in female cynomolgus monkeys (8).…”

“…Inflammation facilitates that infiltrative process and thus would be expected to be associated with the severity of the structural change. 18,19 Nonetheless, inflammation without endothelial dysfunction might not be associated with atherosclerosis, and endothelial dysfunction without inflammation might have a more benign course. Thus the correlations among these markers -inflammation as identified by CRP, endothelial dysfunction as identified by reduced small artery elasticity, 20 and structural changes in the conduit arteries as identified by large artery stiffness -would be expected to be modest.…”
