Direct from solid natural products to pure compounds in a single step: Coupling online extraction with high‐speed counter‐current chromatography

Abstract: Extraction is the most important step in the purification of bioactive compounds from natural products. This study introduces a simple online extraction strategy coupled with high-speed counter-current chromatography for efficient extraction and purification of bioactive components from solid natural products. For online extraction strategy, 1.0 g of ground Mangnolia officinalis or Piper nigrum was loaded into a guard column, which was then positioned on the manual injection valve instead of the sample loop. “…As a perspective, the implementation of such sorbent to on‐line SPE coupled to HPLC‐UV or 2D‐HILIC/RPLC followed by automated fraction collection to Rhinella marina (L.) venom extracts should enable to isolate promising pharmacologically active compounds with higher throughput and purity rates and on a bigger scale. Other techniques to isolate compounds from natural sources could also be considered, such as counter‐current chromatography, extraction on macroporous resins combined to preparative HPLC .…”

“…Natural products have been used in medical purposes for hundreds of years, which are good exogenous candidates (e.g. antioxidant, anti‐inflammatory, antimicrobial, and antitumor drugs) [1]. However, natural products usually contain a large number of compounds, and the traditional bioassay‐guided procedure for purification of bioactive compounds is labor and resource intensive [2].…”

“…HPLC–QTOF‐MS/MS has been widely used for identification of bioactive compounds with high separation resolution, unambiguous precursor, and product ions information [1,4]. Nevertheless, produced massive MS data make manual annotation a rather tedious and difficult work [4].…”
