Differentiation of a Fibrin Gel Encapsulated Chondrogenic Cell Line

Abstract: Hyaline cartilage has very limited regenerative capacity following damage. Therefore engineered tissue substitutes have been the focus of much research. Our objective was to develop a fibrin-based scaffold as a cell delivery vehicle and template for hyaline cartilage regeneration, and compare its cellular properties against monolayer and pellet culture for chondrogenic cells. The chondrogenic precursor cell line, RCJ 3.1C5.18 (C5.18), was chosen as a test system for evaluating the effect of various culture con… Show more

“…There have been reports of cartilage-like tissue formation when uncross-linked fibrin hydrogels were used for tissue engineering of articular cartilage [Peretti et al, 2006;Dare et al, 2007]. Several studies used commercially available fibrin glue of bovine origin for gel formation [Mouw et al, 2005;Park et al, 2005].…”

“…Fibrin is another naturally occurring material that has shown promise in recent years as a cell delivery vehicle [122,185] and injectable scaffold. [104] The main advantage of fibrin gels is that fibrinogen can be obtained autologously from the plasma, thereby reducing the risks of a foreign body reaction.…”

“…The comparative C T method 36,37 was used to analyze the results. Chondrocytes from the same patient and gel type at day 1 were used as a baseline for each gene, unless otherwise indicated, which was normalized to GAPDH housekeeping expression.…”
