Differential Proteome Analysis of Hybrid Bamboo (Bambusa pervariabilis × Dendrocalamopsis grandis) Under Fungal Stress (Arthrinium phaeospermum)

Abstract: In this study, TMT (tandem mass tag)-labeled quantitative protein technology combined with LC–MS/MS (liquid chromatography-mass spectrometry/mass spectrometry) was used to isolate and identify the proteins of the hybrid bamboo (Bambusa pervariabilis × Dendrocalamopsis grandis) and the bamboo inoculated with the pathogenic fungi Arthrinium phaeospermum. A total of 3320 unique peptide fragments were identified after inoculation with either A. phaeospermum or sterile water, and 1791 proteins were quantified. A to… Show more

“…Pathogen spore solution was then injected three times, 30 μL each time, with an interval of three days (day 3, 6, and 9), and cultured in a greenhouse at 28 ± 2°C to investigate the incidence rate of corm rot. The blank group was injected with PDB or NB, and the positive control group was injected with carbendazim, 12 corms per group, repeated three times ( Li et al., 2019 ).…”

“…Most of the studies on the disease are about the pathogen A. phaeospermum , including the isolation and identification of the pathogen, the characteristics of disease occurrence, prevention methods, and other aspects of its physiological biology [ 7 , 8 , 9 ]. Moreover, studies on this pathogen also include bioinformatics such as whole-genome, transcriptome, protein analyses, and molecular biology analysis, such as the functional verification of virulence-related genes of the pathogen [ 10 , 11 , 12 ]. The research on the host B. pervariabilis × D. grandis has only involved proteome and transcriptome sequencing analysis after the plant has been infected by the pathogen [ 13 , 14 , 15 ].…”

“…The differentially expressed proteins were mainly concentrated in biological processes, and the four differentially expressed proteins, sucrose synthase, adenosine triphosphate citrate synthase β chain protein 1, peroxidase, and phenylalanine ammonia lyase interacted significantly with a variety of proteins. The metabolites in response to pathogen infection are mainly concentrated in metabolic pathways, such as the citric acid cycle and phenylpropane biosynthesis [ 31 , 32 , 33 ]. However, this is only an independent analysis of the changes in the expression of host plant genes, proteins, and metabolites under specific pathological conditions, and there is a lack of research on the overall changes in gene expression in the process of pathogen–host interaction.…”
