Differential Expression of miR-499 and Validation of Predicted Target Genes in the Testicular Tissue of Swine at Different Developmental Stages

Abstract: microRNAs (miRNAs) represent a newly identified class of nonprotein-coding *22 nt small RNA that plays important roles in multiple biological processes by degrading targeted mRNA or repressing mRNA translation. This study observed the morphology of swine testicular tissue at different developmental stages (including 1-day old, 1-7 month old) by Hematoxylin-eosin staining. We also examined the expression of miR-499 and its target genes (CYLC1, DMRT1, QKI, XRN2, ZNF313) in samples of tissue slices using quantita… Show more

“…miR-375 has been suggested to play an important role in testicular differentiation in Xenopus [ 70 ]. As in tilapia, miR-499 is also a dominant miRNA in swine, indicating its involvement in testicular development [ 71 ]. miR-9 and miR-192 are also upregulated in tilapia ovary, while miR-27d, miR-29b, miR-92b, miR-144-5p and miR-455 show the same pattern of greater expression in testes at later stage [ 43 ], suggesting they might play crucial roles in tilapia sex differentiation.…”

“…QKI can express four subtypes during the development of chicken sperm (Mezquita et al 1998), and quaking (qkv mutant) mice also show severe https://doi.org/10.17221/158/2018-CJAS sperm maturation disorders. In addition, the study showed that QKI expression is increasing in 1-to 7-month-old boar's testes (Zhang et al 2015). These results indicate that QKI plays an important role in the development of testes and in spermatogenesis.…”

“…Our previous study and that of others showed that miR-499 exhibits a higher expression level during the fetal and prepubertal periods than in the pubertal period in porcine testicular tissues (Ran et al 2015. It was further reported that miR-499 targets the QKI gene and represses its expression in immature porcine Sertoli cells (Zhang et al 2015). Therefore, we speculated that miR-499 might play regulatory roles in immature porcine Sertoli cell growth.…”
