Differential DNA Damage Induced by Chemical Mutagens in Cells Growing in a Modified Selye’s Granuloma Pouch

Abstract: Rapid growth of granulation tissue was induced in rats by injection of croton oil into a subcutaneous air pocket. Growth characteristics of the granulation tissue were evaluated by histopathologic techniques and measurement of 3H-thymidine incorporation into DNA. The DNA of cells growing in the granuloma was labeled with 3H-thymidine. Subsequently, 4 classes of test chemicals (monofunctional and polyfunctional alkylating agents, DNA intercalating agents and chemicals not known to interact… Show more

“…Vasantha (16) reported the inhibition of nucleic acid polymerases in parthenintreated FleLa cells. In addition to the above mechanisms of cytotoxicity, evidence is also accumulating in support of the potent alkylating nature of sesquiterpene lactones that could result in subsequent demage to DNA per se (7,8,(38)(39)(40)(41)(42). Recently, Lion et a!.…”

“…After an expression time of suitable duration granuloma cells are isolated and can be assayed for the presence of DNA single strand breaks (Lee and Zbinden 1979), chromosome aberrations (Maier, Send offprint requests to G. Zbinden at the above address 0340-5761/80/0045/0227/$ 01.20 unpublished observation), sister chromatide exchanges (Maier, unpublished observation), and for the presence of point mutations, e.g., ouabain resistance (OUA R, Maier et al 1978), and 6-thioguanine resistance (HGPRT-, Maier et al 1980). The target cell population consists of fibroblasts growing on the inside of a subcutaneous (s.c.) air pocket.…”
