Differential Antioxidant Responses and Perturbed Porphyrin Biosynthesis after Exposure to Oxyfluorfen and Methyl Viologen in Oryza sativa

Abstract: We compared antioxidant responses and regulation of porphyrin metabolism in rice plants treated with oxyfluorfen (OF) or methyl viologen (MV). Plants treated with MV exhibited not only greater increases in conductivity and malondialdehyde but also a greater decline in Fv/Fm, compared to plants treated with OF. MV-treated plants had greater increases in activities of superoxide dismutase (SOD) and catalase (CAT) as well as transcript levels of SODA and CATA than OF-treated plants after 28 h of the treatments, w… Show more

“…Metabolites in the porphyrin biosynthetic pathway are highly photoreactive and can easily be excited and transfer the energy to O 2 . Proto IX and protochlorophyllide (Pchlide) are especially reactive with O 2 to form reactive oxygen species (ROS), which is harmful to cells and causes the peroxidation of membrane lipids [3,4]. …”

“…For native staining, the antioxidant enzymes (30 µg) were electrophoresed in 10% resolving and 4% stacking gel, respectively, for APX and CAT isozymes, whereas, 15% resolving gel and 5% stacking gel were used for separating the SOD isozymes at 4 • C for 4 h at 80 V in a Tris-Glycine (pH8.3) running buffer. The active staining of isozymes of SOD, CAT, and APX were performed according to Pham et al [34].…”

“…In most cases, protoporphyrinogen oxidase is the last enzyme before the branch in the porphyrin biosynthetic pathway catalyzes the oxidation of protophorinogen IX to Protoporphyrin IX (Proto IX). Protophorinogen IX is then directed to the magnesium (Mg) and iron (Fe) branches for chlorophyll and heme biosynthesis, respectively [ 16 , 17 ].…”
